Objective:We evaluate the expression of PTEN, HIF-1α and MMP-9 protein in colorectal carcinoma tissue, and their invasion and metastasis.Find out the objective idicators and possible mechanisms of the occurrence, development,invasion and metastasis between those three factors and colorectal cancer.Method: Check the conditions of Pten, HIF-1αand MMP-9 in 56 cases of colorectal cancer tissues and 24 cases of normal tissue which are all pathologically confirmed by Immunohistochemical SP method(streptavidin avidin peroxidase immunohistochemistry).Data results use SPASS19.0 statistical package for statistical analysis, and correlation analysis uses spearman rank correlation analysis,Meanwhile, calculate the survival time based on the patient’s follow- up data,Analyze and compare the three factors with the clinical stage, histological grade, depth of invasion,lymph node metastasis and prognosis in patients who got colorectal cancer,and discuss its clinical significance.Result: 1.The positive expression number of PTEN protein in colorectal carcinoma was 18(32.1%). In normal tissues, the positive expression number was 15(62.50%). The positive expression number of HIF-1a protein in colorectal carcinoma was 40(61.7%). In normal tissues, the positive expression number was 10(41.66%).The positive expression number of MMP-9 protein in colorectal carcinoma was 29(51.7%). In normal tissues, the positive expression number was 6(25.0%).The positive expression rates of PTEN,HIF-1a and MMP9 protein in colorectal carcinoma was significantly higher than in adjacent normal tissue,the difference was significantly different(P <0.01).2.The expression levels of PTEN,HIF-1a and MMP9 have correlation with tumor invasion, lymph node metastasis and Dukes stage(P <0.05),but have no correlation with the patient’s age, sex, tumor location, size differentiation(P> 0.05). 3.The correlation in the expression of PTEN,HIF-1a and MMP9:In the result of analysis by Spearman Test,the expression of 2 shows significantly positive correlation with that of MMP9(rs=0.339,p=0.011),while the expression of HIF-1a shows significantly negative correlation with that of PTEN(rs=-0.580,p=0.001).The correlation between the exoression of PTEN and MMP9 was negative but not signficently(rs=-0.407,p=0.002).The result showed that those three proteins were correlated with each other in human colorectal carcinoma.The abse nce of PTEN enhanced the transcription of MMP9,MMP9 may be the primary downstream genes of HIF-1a in colorectal carcinoma. The patients’ survival time with the positive expression of Her-2,Stat3 and MMP-9 is less than negative ones(P<0.05).Conclusion: 1.According to this lab data,We can speculate tha Pten,HIF-1a and MMP-9 correlate significantly and play a key role in colorectal carcinoma occuring,developement,metastasis and prognosis.Advanced research has shown that PTEN,HIF-1a and MMP-9 protein help somewhat in indicating colorectal carcinoma.Their expressions are related to part of colorectal carcinoma clinic pathologic characters and worthy in clinic judge of colorectal carcinoma prognosis and advanced therapy.In colorectal carcinoma tissues,HIF-1a expression in correlate significantly positive with MMP-9 expression and PTEN is correlate significantly negative with HIF-1a expression.The negative correlation between PTEN and MMP-9 expression shows PTEN may adjust negatively HIF-1a expression and depress MMP-9 expression though PI3K/AKT signal transfer passage. 2. Combined detection of three indicators to judge the invasion, metastasis and prognosis of colorectal cancer may have important guiding significance.
