Abstract/Summary:
ObjectivesTo observe the effect of electroacupuncture at Minngmen point on OPG-RANKL signaling of femur in postmenopausal osteoporosis rats.Methods4.5 month female SD rats were randomly divided into sham operation group (Sham), model group (Ovx), electroacupuncture Mingmen group (GV4), electroacupuncture non-acupoint group (Mock). Except for the sham group, the rats were performed ovariectomized (ovariectomy, Ovx) to induced the model of postmenopausal osteoporosis. After the models were prepared, rats from different group were performed relatively treatment. After 3 courses of treatment, bone miner density, biomechanics and OPG-RANKL signaling by different method.Results1. Biomechanical index and bone miner density:Compared with Ovx group, Mingmen group can significantly improved the femur fracture load and the maximum load as well as the bone miner density (P<0.05); There was no significant change between the Mingmen group and the sham group (P> 0.05) statistically; There was no significant change between the Ovx group and the mock group (P> 0.05).2. Expression of OPG and RANKL in different experimental groups: Immunohistochemical results showed that expressions of OPG and RANKL were significantly reduced in rats in the Ovx group compared with those in rats in the sham group (P<0.05) and this reductions were significantly reversed by electroacupuncture at Mingmen group (P<0.05), but not at non-acupoint (P>0.05).ConclusionElectroacupuncture at Mingmen point can significantly improved the femur fracture load and the maximum load as well as the bone miner, and activing the OPG-RANKLsignaling pathway might be one of the mechanism by which the electroacupuncture at Mingmen point treating the postmenopausal osteoporosis.
