Influence Of Fufang Sanqi Xiaotong Ointment Phonophoresis On MMP-13 In Chondrocytes Of Rabbit Knee Osteoarthritis

Objective:To explore the curative effect of fufang sanqi xiaotong ointment phonophoresis, ultrasound and fufang sanqi xiaotong ointment outside treatment on rabbits knee osteoarthritis, and the influence on chondrocytes MMF-13 expression.Methods:Thirty New Zealand white rabbits were randomly divided into five groups: phonophoresis group, ultrasound group, ointment group, model group and normal group. In addition to the normal group, the other four group rabbits received anterior cruciate ligament transaction to make animal models of KOA, and the intervention carried out after six weeks. The phonophoresis group and ultrasound group used ultrasonic therapy equipment to treatment, by used the intensity of 300 mW/cm2, pulse on-off ratio is 20% and frequency of 1 MHz. The ointment group and model group also used ultrasonic therapy equipment to treatment, but the instrument is not plugged. Treatment once a day,10 minutes at a time, and six days’ treament is a course, there are two courses totally. Remove the femoral condyle cartilage after two weeks’ intervention. Hematoxylin-eosin staining was conducted for histological observation of rabbit articular cartilage, and the cartilage structure were assessed and quantified with the modified Mankin rating criteria, immunohistochemistry were used to assess the expressions of MMP-13 in rabbit chondrocytes.Results:1. Comparing with the normal group, the treated groups and model group showed different degree of osteoarthritis. The phonophoresis group, ultrasound group and ointment group showed lighter rabbit knee osteoarthritis cartilage damage compared with the model group.2. Hematoxylin-eosin staining showed that the articular cartilage pathological changes of treated groups were lighter than model group, and the phonophoresis group was the lightest in the treated groups. Comparing with the normal group of the modified Mankin score, the treated groups and model group showed higher scores(P<0.05); Comparing with the model group of the modified Mankin score, the treated groups showed lowed scores(P<0.05), the score of ultrasound group was lower than ointment group, but there was not significant difference(P>0.05).3. Immunohistochemistry staining of MMP-13 showed very low expression level of normal group, and the model group rabbit chondrocytes showed significant high expression level of MMP-13. Comparing with the normal group, the treated groups and model group showed higher expression level of MMP-13(P<0.05); Comparing with the model group, the treated groups showed lower expression level of MMP-13(P<0.05); Comparing with the phonophoresis group, the ultrasound group and ointment group showed higher expression level of MMP-13(P<0.05); The ultrasound group showed lower expression level of MMP-13 than the ointment group, but there was not statistical significant between them(.P>0.05).Conclusion:1. Fufang sanqi xiaotong ointment phonophoresis, ultrasound and fufang sanqi xiaotong ointment outside treatment can effectively delay the rabbit knee osteoarthritis cartilage damage, and fufang sanqi xiaotong ointment phonophoresis has a better curative effect than ultrasound therapy alone or fufang sanqi xiaotong ointment outside treatment.2. The therapeutic mechanism of fufang sanqi xiaotong ointment phonophoresis, ultrasound and fufang sanqi xiaotong ointment outside treatment may be by inhibiting the MMP-13 expressing of rabbit chondrocytes.
