Experimental Research On The Changes Of Hemorheology In Early NAFLD Rats And Its Relationship With Blood Stasis Syndrome

Objective: To observe the dynamic changes of hemorheology in rats with early nonalcoholic fatty liver:to study the relationship between hemorheology and blood stasis syndrome with early nonalcoholic fatty liver disease.Methods:I.Preparations:160 healthy and male rats were randomized into 5 groups, normal group with 50 rats, model group with 50 rats, Gexia Zhuyu Decoction group with 20 rats, Simvastatin group with 20 rats, Aspirin group with 20 rats. The normal group were fed with normal animal forages,while the other four groups were fed with high fat forage(adding 1 0%of lard oil and 2%of cholestenone to normal animal forages) for 12 weeks.II.Group administration: at the end of the 8th week rats in the model group and the normal group were given gavage of physiological saline and rats in Gexia Zhuyu Decoction group, Simvastatin group and Aspirin group respectively were given gavage of Gexia Zhuyu Decoction soup, Simvastatin and Aspirin, 1ml/200 g weight, 1 time per day. III.Index detecting :at the end of the 4th, 6th,8th, 10 th and 12 th week, taking the blood sample and liver tissue sample from rats in each group respectively,testing the weight of rats,the wet weight of liver weight/body weight, histopathological change of pathological section of liver tissue after hematoxylin-eosin staining with microscope, blood fat: TC and TG, liver fat: TC and TG, hemorheology: whole blood viscosity,plasma viscosity, red cell assembling index, red cell deformability index, plasma fibrinogen and platelet aggregation rate. Results: I.General situation: rats in the normal group and the model group eating well from the start to the weekend of the 4th 0f experiment, compared to the normal group, the model group rats were higher in feed-intake and weight increasing at the weekend of the 4th, the model group rats were obvious obesity and drumble at the weekend of the 8th,statistcs found out: compared to the normal group,weight,liver wet weight and liver weight/body weight of the model group rats were significantly higher(P<0.05 or P<0.01) at the end of the 4th,6th,8th, 10 th and 12 th week. II Morphologic changes: liver tissue of the normal group rats with naked eyes showed: bright red, the edge is sharp, good elasticity. Pathological section of liver tissue after hematoxylin-eosin staining and Sultan III staining with with light microscope showed that hepatic cord radially distributed along the central vein, liver cell without hydropic degeneration and fatty degeneration at the end of the 4th, 6th,8th,10 thand 12th week. At the end of the 4th week, liver tissue of the model group rats with naked eyes showed: liver tissue morphology and color was normol, at the end of the 6th week, liver volume of the rats in the model group increased, at the end of the 8th week, compared to the normal group, liver volume of the rats in the model group increased significantly, having the feeling of greasy when touch the section. Observed with light microscope showed that: 1/3 more fatty degeneration of liver cells.At the end of the 10 th and 12 th week fatty degeneration of liver cells of the model group rats gradually increased,at the end of the 12 th week 2/ 3 more fatty degeneration of liver cells.III.Biochemistry index: compared to the normal group, Serum TC,liver TC and liver TG had significant different(P<0.01) at the end of the 4th,6th,8th,10 th and 12 th week. Serum TG increased significantly(P<0.05或P<0.01)at the end of the 6th,8th,10 th and 12 th week.Compared to the model group, Serum TC,Serum TG of rats in Gexia Zhuyu Decoction group, Simvastatin group Significantly decreased(P<0.05或P<0.01)at the end of the 10 th and 12 th week, Compared Gexia Zhuyu Decoction group with Simvastatin group, there had no significant difference between the two groups(P>0.05). Liver TC and liver TG of rats in Gexia Zhuyu Decoction group, Simvastatin group decreased:liver TG of rats in Gexia Zhuyu Decoction group Significantly decreased(P<0.01),and liver TC and liver TG of rats in Simvastatin group Significantly decreased(P<0.05) at the end of the 10 th week,liver TG of rats in Gexia Zhuyu Decoction group Significantly decreased(P<0.05) at the end of the 12 th week, Compared Gexia Zhuyu Decoction group with Simvastatin group, there had no significant difference between the two groups(P>0.05). IV.Hemorheology index:at the end of the 4th week hemorheology index of rats in the model group close to the normal group(P>0.05). Whole blood viscosity, plasma viscosity, red cell deformability index, plasma fibrinogen, platelet aggregation of rats in the model group increased significantly(P<0.05或P<0.01)at the end of the 6th,8th,10 th,12th week, red cell assembling index of rats in the model group increased significantly(P<0.05或P<0.01)at the end of the 8th,10 th,12th week.Compared to the model group, whole blood viscosity, plasma viscosity, red cell assembling index,plasma fibrinogen, platelet aggregation of rats in Gexia Zhuyu Decoction group and Aspirin group decreased significantly(P<0.05或P<0.01)at the end of the 10 th week,red cell deformability index of rats in Gexia Zhuyu Decoction group and Aspirin group decreased significantly(P<0.05或P<0.01)at the end of the 12 th week. Compared Gexia Zhuyu Decoction group with Simvastatin group, there had no significant difference between the two groups(P>0.05). Conclusion: I. The occurrence and development of early nonalcoholic fatty liver disease be related to hemorheology and blood stasis syndrome; the index of hemorheology can be used the reference index for the diagnosis and curative effect of blood stasis syndrome of early nonalcoholic fatty liver disease. II.Gexia Zhuyu Decoction could also decrease serum TC and serum TG, liver TC and liver TG in early nonalcoholic fatty liver rats.
