Objective:Diallyl trisulfide is the chemical name of Garlicin,it is a chemical compound separate from garlic corm. There are some reports that garlic could treat ileus as a Chinese drugs agentia. In recent years, there are some literatures about the acceleration effect of garlicin on gastrointestinal function, but dimness on mechanism. Motilin is one of the of brain-gut peptide to activate gastrointestinal motility, it is act on motilin receptors in gastrointestinal nervous systerm and smooth muscle cell, and then initiate Migrating motility complex. Ghrelin combine with its receptors, then initiate in gastric emptying on central and peripheral pathway. Ghrelin is also one of the of brain-gut peptide to activate gastrointestinal motility. This study is to examine the effect of garlicin, motilin and ghrelin on gastrointestinal function in mice.Methods:90 mice are adaptive feeding one week in animal room with photoperiod 12/12> 25℃、humidity 60%, free feed by Lanzhou university animial centre.The mice were randomly divided into high-dose-garlicin, middle-dose-garlicin, low-dose-garlicin group, metoclopramide group and control group. These mice with no food and drink for 24 hours, all of mice were divided with 50 mg/kg of garlicin to high-dose-garlicin group,20 mg/kg of garlicin to middle-dose-garlicin group,5 mg/kg of garlicin to low-dose-garlicin group,8 mg/kg of metoclopramide to metoclopramide group and 10 mg/kg of saline to saline control group by subcutaneous injection.0.4 ml 2% dextran blue 2000 was in tragastrical after 30 min. Twenty minutes later, mice were sacrificed after taking blood. According to anatomy principle of mice, ligating gastric cardia and pylorus, dissociating stomach and small intestine, measuring the rate of gastric emptying and small intestinal propulsion, After prepare the mice serum,measuring the motilin and ghrelin concentration by ELISA respectively.Results:1. Compare with the control group (74.17± 10.23%), gastric emptying rate in high-dose-garlicin group (83.86±8.19%), middle-dose-garlicin group (86.99±7.57%) and metoclopramide group (81.74±6.83%) are significantly higher,with a statistical significance (P<0.05); No statistical significance (P>0.05) in low-dose-garlicin group and control group; Compare with the metoclopramide group, gastric emptying rate in high-dose-garlicin group, middle-dose-garlicin group and low-dose-garlicin group with no statistical significance (P>0.05)2. Compare with the control group (37.52±9.57%), small intestinal propulsion rate in high-dose-garlicin group (60.28±7.12%) and middle-dose-garlicin group (53.49±8.35%) are significantly higher, with a statistical significance (P<0.05), same in metoclopramide group (43.37±13.92%);There are no statistical significance (P>0.05) on small intestinal propulsion rate compare with low-dose-garlicin group, metoclopramide group and control group.3. Compare with the control group (154.99 ± 66.47 ng/L), motilin consistence in high-dose-garlicin group (220.53±58.32 ng/L) and middle-dose-garlicin group (275.26±57.85) are significantly higher, with a statistical significance (P<0.05); Compare with the metoclopramide group (65.82±80.56 ng/L), motilin consistence in high-dose-garlicin group and middle-dose-garlicin group are significantly higher, with a statistical significance (P<0.05); Compare with the control group, no statistical significance (P>0.05) in low-dose-garlicin group and metoclopramide group.4. Compare with the control group (5.36±3.06 ng/ml), ghrelin consistence in high-dose-garlicin group (12.38±3.29 ng/ml) and middle-dose-garlicin group (15.63±5.38 ng/ml) are significantly higher, with a statistical significance (P<0.05); Compare with the metoclopramide group (7.74±5.92 ng/ml), motilin consistence in high-dose-garlicin group and middle-dose-garlicin group are significantly higher, with statistical significance (P<0.05); Compare with the control group, no statistical significance (P>0.05) in low-dose-garlicin group and metoclopramide group.Conclusions:Garlicin significantly promoted the secretion of motilin and ghrelin, then gastric emptying and small intestinal propulsion rate also increased.
