1、Backgroung and purposeMiRNAs are small non-coding RNAs that negatively regulate gene expression at the post-transcriptional level. Plenty of earlier studies have shown that miRNAs is frequently differentially expressed in many human tumors and they can function as either oncogenes or tumor suppressor. The miRNAs could be utilized as biomakers for diagnosis and prognosis prediction of cancers if we know the mechanism of it’s. A recent study has found that the expression lever of mir-424 in hepatocellular carcinoma tissue is lower than the normal tissue. So, we could use the gene transfer techniques to transiently transfecte the mir-424mimics into human liver cancer cells MHCC97-H and MHCC97-L, than we can investigate the effects of mir-424 on proliferation, migration and invasion of human liver cancer cells. We try to explore the mechanism of mir-424 in liver cancer cells preliminarily.2、MethodThe expression lever of mir-424 in human liver cancer cells MHCC97-H, MHCC97-L and hepatocyte HL-7702 were assessed by real-time qRT-PCR. Then the chemicalsyn thesis of mir-424mimics was transiently transfected into human liver cancer cells MHCC97-H and MHCC97-L respectively. Furthermore, the change of mir-424 expression lever was detected by real-time qRT-PCR. Finally, the cell proliferation was detected by MTT and cell migration and invasion were detected with the Transwell test.3、Result1) Compared with hepatocyte HL-7702, the expression lever of mir-424 was remarkably down-regulated in liver cancer cells MHCC97-H and MHCC97-L;2) The real-time qRT-PCR experiments, the expression lever of mir-424 was remarkably up-regulated in liver cancer cells MHCC97-H and MHCC97-L after transfected by mir-424mimics.3) MTT assay showed that liver cancer cells MHCC97-H and MHCC97-L with over-expression of mir-424 are more likely to have poor proliferation in contrast to control groups.4) Transwell test revealed that the migration and invasion of liver cancer cells MHCC97-H and MHCC97-L with over-expression of mir-424 were obviously inhibited in contrast to the control groups.4、ConclusionsExperimental investigation shows that, the proliferation, migration and invasion in human liver cancer cells MHCC97-H and MHCC97-L will be inhibited when they over-expression with mir-424 through gene transfer techniques in vitro.
