Objective:Vanillin is used very widely. For example it is used in daily chemical production, food, the tobacco industry and manufacture of clinical medicine. And at the same time, Vanillin is a very important spice. It is often used as food flavor to allocate in butter, milk, chocolate, cocoa, yogurt, butter, rum, vanilla, pineapple, peach, banana, walnut, strawberry, cherry, melon, etc. It is often used for food additive and joined into the biscuits, Chocolate, milk tea, ice cream, chewing gum, candy, and pastries. Human infertility rate are increasing year by year, and it is related to many factors, such as Environment, food and drugs. So far, there are no relevant reports about the affect of vanillin on human reproductive system in China. From the experiment which of the male sperm vitality detects the effect of vanillin on the grade A and A+B grade of the male sperm vitality, it can be to view the food additive that vanillin has any impacts on male infertility.Methods:The specimens come from the men who are in the first affiliated hospital of Lanzhou university reproductive specialized subject hospital concentration where the infertility couples received the assisted reproductive technology. Specimens are in the normal range which WHO sets. The standard is that:No serious reproductive system infection recently; no hormonal drug taking history; no serious body trauma and family genetic disease history; chromosome is normal:46, xy; no affected by X-ray and other radiation in the half past year; no abnormalities of reproductive system in the physical examination. The specimens of patients are informed to signed consent before the experiment and through the ethics committee’s approval. The WHO formulate the normal reference range of semen specimen:The color is gray, or light yellow; volume 3=2.0 ml; (2) pH≥7.2; (3) density≥20×106ml; (4) sperm motility:A+B≥50%(and a≥25%). Add sperm to the EP tube after fluid collection in half past hour. Put it in the constant temperature of 36.8 ℃ box to liquefy for 30min. Gently, slowly blowing and blending each specimen. The concentration of vanillin is divided into different groups:1.0×10-3 mmol/ml, 1.0×10-4 mmol/ml, 1.0×10-5 mmol/ml,1.0×10-6 mmol/ml,1.0×10-7 mmol/ml. And then they were added to the normal semen. Tested and compared their influence on sperm vitality through computer assisted sperm analyzer. (CASA)Results:As the vanillin concentration decreasing, its effect is more and more weak on sperm motility. The result is more and more obvious.The change is minimal on the concentration of 1.0×10-6 mmol/ml. And there are no significant differences compared with normal saline and water sterilization. There is no statistical significance after the mathematical statistical analysis.Conclusion:Vanillin is a kind of relative safety and used widely food additive. There is no influence sperm motility if it was used within the scope of the provisions of the state standard. Do not result in lower sperm motility, and affect infertility which was caused by male factor. But the other more effects on human remains to be explored.
