Objective: To investigate the effects of panax notoginseng saonins(PNS) on inflammatory mediators of cardiopulmonary bypass caused to brain damage in rats. Methods: Select 60 adult male SD rats, 30 used in CPB surfery for blood, the other 30 were randomly divided into three groups, the normal control group, the CPB group(CPB time 60 min), and the PNS group:panax notoginseng saonins drug+CPB group. The latter according to different PNS duration is divided into three subgroups: the group of drug use to intravenous infusion pump for input in surgery, the group of drug use to CPB prefilled liquid, the group of drug use to transvenous one-time treatment after anesthesia the rat. In addition to the normal control group, take three SD rats in the rest of the group respectively before CPB, CPB stop instantly, CPB surgery 6 hours, 12 hours after CPB, 24 hours after CPB five point extraction femoral artery of rats with ELISA method line plasma TNF alpha, IL-6, IL-1beta content detection; there’s only three SD rats in 24 hours after CPB in brain tissue for immunohistochemical detection line ICAM-1. Results: CPB group compared with normal group, the serum TNF alpha, IL-6, IL-1 beta content is significantly higher than normal group(p<0.05), this suggests that the extracorporeal circulation can lead to the content of related inflammatory cytokines in plasma of rats increased significantly. Treatment group compared with CPB group down immediately, the only V group of TNF alpha, IL-6, IL-1beta levels were significantly reduced(P<0.01), the rest of the no difference between the treatment group. Explain anesthesia in rats after transvenous one-time treatment effect of treatment group was obviously higher than that of the rest of the two groups. Conclusions: In extracorporeal circulation in rats, the early use of PNS can inhibit the expression of TNF alpha and IL-1beta, reduce TNF alpha and IL-1beta signaling, greatly improve the cell oxygen free radical removal function, inhibits the activation of inflammatory white blood cells, decrease the release of IL-6,ICAM-1, hindered the adhesion and accumulation of neutrophils, thereby reducing the blood platelet and endothelial cell adhesion, reduces the chemotactic movement, the protection of brain play a role.
