Objective:1.To study on the chemical components of EtOAc fraction and n-butanol fraction of Korla fragrant pear.2.1To confirm the extraction processes of total flavone and polysaccharides from Korla fragrant pear fruit juice and peels.The content of total flavone and polysaccharides were determined.3.To study on the anti-oxidation activities of total polysaccharides from Korla fragrant pearATo explore the antitussive, expectorantand immunoregulation effects of crude polysaccharides from Korla fragrant pear.Method:1.The chemical components of EtOAc fraction and n-butanol fraction were extracted and isolatedby recrystallization with solvent and silica and glucan gel column chromatography, and the structures of them identified by theirspectroscopic datas(UV,1H-NMR,13C-NMR).2.Used the L9(34) orthogonal design to arrange experiment at the base of the single factors experiment’s foundation,and total flavone and total polysaccharides content determined by the ultraviolet spectrophotometr y,moreover,ensuring the best extraction processes.3.To evaluate the ability of removal to DPPH·,·OH,-O2- and ABTS+,as well as reducing iron ion of polysaccharides from Korla fragrant pear with the ultraviolet spectrophotometry.4. Observedthe effective of crude polysaccharides from Korla fragrant pear in antitussive, expectorant.Furthermore,toexplore theimmunoregulation effects of crude polysaccharides with IL-2, IFN-γ and the phagocytic index of macrophage as observation indexes.Results:1.Six compounds were isolated from Korla fragrant pear.The compound I(oleanolic acid),Ⅱ(arbutin),Ⅲ(β-sitosterol) IV(quercetin)were isolated from EtOAc fraction.The compound V(methyl oleanolate),VI(ursolic acid),VIwere isolated from n-butanol fraction.2. ① Ultimately to confirm the best iextraction processes of total flavone from Korla fragrant pear fruit juice for:50% concentration of alcohol,40℃ for 120minutes, total only times.The best extraction processes of total flavone from Korla fragrant pearpeels for:85% concentration of alcohol,40℃ for 60 minutes,total second times.Under the condition,the a only once verage yield of total flavone from Korla fragrant pear fruit juice and peels respectively is 1.79mg/g and 5.12mg/g.②Ultimately to confirm the best extraction processes of polysaccharides from Korla fragrant pear fruit juice for:the ration of solid material to liquid is 1:40,60℃ for 1h, only once. Under the condition,the average yield of polysaccharides of Korla fragrant pear fruit juice respectively is 78.29mg/g.3.The crude polysaccharides from Korla fragrant pear have the good anti-oxidation ability:it has the ability of removing DPPH-,OH,·O2-,ABTS+,and effectively reducing the iron ionAThe polysaccharides from Korla fragrant pearwere able to prolong the cough latent period, reduce the frequency of cough and increase the phlegm expelling.The three doses polysaccharides group of Korla fragrant pear all have the functions of increasing the content of IL-2 and IFN-y of mice, and increasing the weight of mice thymus and spleen, and increasing phagocytotic ability of macrophage.Conclusion:1.Six compounds were isolated from Korla fragrant pear,2.It has been proved that the determination is convenient, and the result isstable under the result of extraction processes experiment of orthogonal design.It is a pretty method to use determining total polysaccharides and total flavone.3.The crude polysaccharides from Korla fragrant pearhave the good anti-oxidation ability.Crude polysaccharides from Korla fragrant pearalso were able to promote the secretion of IL-2 and IFN-y of immunocompromised mice, as well as enhance the organism index and phagocytic index. It has proved that crude polysaccharides from Korla fragrant pear have the functions of antitussive, expectorant and immunological enhancement.The results of this study are warranted to further study on the pharmacological effects of Korla fragrant pear.
