| Glehnia littoralis.is the dry root of Glehnia littoralis Fr.Schmidt ex Miq.It is a commonly used Chinese herbal medicine and Mongolian medicinal material.It is used for lung heat cough,labor and blood,stomach yin Insufficient,fever,pain,throat and thirst.There are many kinds of sources of G littoralis in the market,but there is no uniform quality control standard,which leads to uneven quality of products.In the2015 edition of the Chinese Pharmacopoeia,only the traits,microscopic identification,sexual taste,and menstruation of G littoralis.were described,while the examination of the medicinal materials and the identification of the extracts,thin-layer chromatography and content determination were missing.The quality control system needs further improvement.In order to further improve the quality control standards of G littoralis.,this study conducts research on the evaluation and material basis of antitussive and expectorant effects,provides a basis for its antitussive and expectorant effect,and establishes a simple,scientific and controllable quality standard system..1.Evaluation of cough and sputum activity of Glehnia littoralis.In this study,the effects of antitussive and expectorant effects on the total extracts of Cordyceps sinensis,its petroleum ether fraction,ethyl acetate fraction,n-butanol fraction and water fraction were studied by using ammonia water cough method and tracheal phenol red excretion method.The results showed that the total extract,petroleum ether fraction and ethyl acetate fraction of G littoralis ignificantly reduced the number of coughs in mice,while the ethyl acetate fraction and n-butanol fraction significantly increased the phenol red excretion in mice,indicating that the north The total extract,petroleum ether fraction and ethyl acetate fraction of G littoralis have obvious antitussive effect,and the ethyl acetate fraction and n-butanol fraction have obvious sputum effect.The reason why the total extract sputum is not obvious may be due to factors such as extraction solvent(ethanol solution),extraction method(heating reflux),and medicinal material origin(Laiyang,Shandong).In addition,in this study,the HPLC fingerprints of the total extracts of G littoralis.and its extracts were analyzed for their correlation with antitussive and antibacterial effects.The results showed that the retention time in HPLC was22.5-40.0min.The chromatographic peak may be the main active component of the antitussive effect of G littoralis.,and the peak of the retention time of5.5-22.5min may be the main active component of the effect of G littoralis..The indicator component has an important guiding role.The results of this experiment show that the water extract(polysaccharide)has no obvious antitussive and expectorant effects.It is suggested that the indicators related to Ziyin tonic,antitussive and expectorant should be selected separately when determining the quality evaluation index of G littoralis.The results of this experiment provide a basis for the clarification of the pharmacodynamics of the antitussive and expectorant of G littoralis.and the selection of quality evaluation indicators.2.Study on chemical constituents of effective parts of cough and phlegm of G littoralis.This study used silica gel,gel,preparative HPLC and other column chromatography to separate and purify,using NMR,UV,IR,MS and other spectral techniques and compared with the relevant literature,isolated and identified from the effective part of G littoralis..Compounds are known.These 16 compounds include threeβ-carboline alkaloids(1-3),four phenylpropanoids(4-7),five phenolic acids(8-12),three polyacetylenes(13-15)and A fatty acid(16).Further systematic chemical taxonomy of the 16 monomer compounds isolated above was carried out.3.Study on quality standard of G littoralis.In this study,the thin-layer chromatography,fingerprint,moisture,ash,leachate,and farainol content of G littoralis.were determined and the corresponding operational specifications were standardized,and the quality standard of G littoralis.was established.Methods:Seven aspects of extraction process and methodological investigation of G littoralis.were studied.The method of TLC identification of Beisha ginseng was established.The fingerprint was prepared by HPLC,and the existing 30 batches of G littoralis.were used.The extraction process and methodological investigation were carried out.The results of the TCM Chromatographic Fingerprint Similarity Evaluation System of the Chinese Pharmacopoeia Committee were used to analyze the results of 30 batches of G littoralis.from different sources and batches.Refer to the Chinese Pharmacopoeia2015 Edition.In the four regulations,the water,total ash,acid-insoluble ash,alcohol extract and water extract of G littoralis.were determined.The content of 30 batches of G littoralis.in different sources was determined by using falcarinol as an indicator.Make the inspection items more complete,more standardized and more scientific.RESULTS:The chemical fingerprints of G littoralis.in different producing areas were established.It can be seen that there are significant differences between the three main producing areas of Hebei,Inner Mongolia and Shandong,and the differences between the same areas are also very different.Significantly;according to the technical requirements of the Chinese Pharmacopoeia medicinal materials quality standards,the quality standards of the northern sand ginseng were established,and the draft quality standard of G littoralis.was formed:(1)Established the TLC identification method of G littoralis.;(2)stipulated the limits of the[check]items of G littoralis.:the water should not exceed 11.0%,the total ash should not exceed 3.9%,and the acid-insoluble ash should not exceed 0.3%.The leachate shall not be less than 10.0%and the water extract shall not be less than 25.0%.(3)Established a method for determination of the content of falcarinol in G littoralis.,and established a limit regulation:This product is calculated on the basis of dry products.and contains not less than 0.05%of falcarinol(C17H24O).(Tare)or 0.11%(not peeled).Conclusion:The above studies provide a scientific basis for the study of the quality standards of t G littoralis. |
