Objective:To study the separation and identification of alkaloid compounds from he leaves of Nitraria sibirica Pall.To determine the optimal extraction condition of Nitraria sibirica seed oil by supercritical CO2 extraction technology and analyze its components by GC-MS. Methods:1.The HSCCC technology were used for the separation of alkaloid compounds from the Nitraria sibirica leaf.2.With oil yield as evaluation index,the orthogonal design L9(34) was applied to optimize supercritical CO2 extraction conditions of Nitraria sibirica seed oil,and fatty acid composition of the oil extracted under the optimal condition was analyze by GC-MS. Results:1.Two new and two known alkaloid monomer compounds separated from the Nitraria sibirica leaf extraction by HSCCC technology and their structure were identified.2. The results showed that the effects of extraction temperature and extraction pressure for the extraction rate of Nitraria sibirica seed oil were higher than the CO2 flow,the optimal extraction conditions were as follows: extraction pressure 40MPa, extraction temperature 55℃, CO2 flow rate10 L/min.Under this conditions, the Nitraria seed oil yield is 29.34%. Among them the unsaturated fatty acid reached to 91.42% mainly contributed by linoleic acid (65.67%) and oleic acid (25.75%),when the saturated fatty acid content mainly contributed by palmitic acid (4.17%) and stearic acid(2.32%).In ddition single unsaturated fatty acid 11-Octadecenoic acid and polyunsaturated fatty acid were rarely reported. Conclusion:This study combined the new efficien separating methods with to modern spectrum analyze methods,separated and purified 4 alkaloid monomer compounds and comfirmed their chemical structures,include two new compounds.This method has the advantages on it’s stability,repetability,retrievability and less sample loss than the traditional separation methods.Overall consideration from the extraction rate, purity,residual solvents of the extraction process,the supercritical CO2 extraction technology is superior to the traditional methods on the extraction of Nitraria sibirica seed oil. It was stable and repeatable.The results of GC-MS analysis of supercritical CO2 extraction of Nitraria seed oil fatty acid composition was expected.This study provide basis for the research of chemical composition of Nitraria sibirica.
