Objective: In this study, we investigated the regulatory effects of craniocerebral firearm injury on terminal ileac Permeability under high tem Perature Plus high humidity, and/or high blood glucose circumstances.Methods: 1. Modeling and grouPs: Following the SOP of craniocerebral firearm injury animal modeling under high tem Perature Plus high humidity, and/or high blood glucose circumstances, 40 healthy male adult mongrel dogs were randomly divided into 8 grou Ps: Normal control grou P(A), high tem Perature and high humidity with high blood glucose grou P(B), craniocerebral firearm injury with normal tem Perature,normal humidity, and normal glucose grou P(C), craniocerebral firearm injury with normal tem Perature, normal humidity, and hy Perglycemia(D), craniocerebral firearm injury with high tem Perature, high humidity,and normal glucose grou P(E), craniocerebral firearm injury with high tem Perature, high humidity, and hy Perglycemia grou P(F), normal tem Perature and humidity with hy Perglycemia grou P(G), high tem Perature and high humidity with normal blood glucose grou P(H). 2.SPecimen collection:. After modeling, dog models were fixed su Pinelyon the o Peration table. In sterile environment, 3-5 cm in the middle of abdomen were cut to ex Pose the ileocecum. 1 to 3cm terminal ileum tissues from 5 dogs Per grou P were cut followed by the anastomosis of the stum P and abdomen. And 3ml Peri Pheral blood sam Ples from each dog were collected at 0/ 0.5/1.0/1.5/2 hours after the surgery. 3. Target Protein assessment: The concentration of D-lactin and diamineoxidase(DAO) in blood sam Ples from dogs at 5 time Points were quantified using ELISA assay, and data in and among grou Ps were com Pared and analysed. Terminal ileum s Pecimens were observed using light microsco Pe to score the damage of teminal ileum mucosa. Score data among different grou Ps at the same time Point were com Pared, and the correlation of it with the concentration of D-lactin and DAO were analysed. The damage levels of mitochondria were scored by electron microsco Pe. Score data at the same time Point were com Pared among different grou Ps, and its correlation with the concentration of D-lactin and DAO were analysed. 4. Biostatistic analysis: All data were analysed by SPSS 18.0 system and Presented as Mean±SD One-way ANOVA and T-test were used for intergrou P data analysis among different grou Ps, and re Peatitive measure ANOVA for intragrou P data analysis(α=0.05). P<0.05 indicates biostastitically significant difference.The study was a PProved by the local ethics committee, and all ex Periments were carried out health advice based on ex Perimental animal care and use of national institutions.Results:1. The alteration of D-lactin concentration in Peri Pheral blood: the concentration of D-lactin in Peri Pheral blood increased in a significant time-de Pendent manner(P<0.05). Also, the levels of D-lactin inuninjured dogs were significantly lower than the craniocerebral firearm injury models at all time Points(P<0.05).Normal tem Perature and humidity grou P in all injury models at varied time Points caused a lower concentration of D-lactin, com Pared with those in High tem Perature Plus high humidity grou Ps(P<0.05). However, no significant changes due to the high levels of blood glucose were detected(P>0.05).2. The alteration of D-lactin concentration in Peri Pheral blood: Similarly,the Protein levels of DAO rised in res Ponse to the extention of time after surgery(P<0.05). At all time Points, uninjured models led to a lower concentration of DAO than the craniocerebral firearm injury models(P<0.05). And along with the injury, normal tem Perature and humidity had a weaker effects on the rise of DAO levels in Peri Pheral blood com Pared to high tem Perature Plus high humidity(P<0.05), while high blood glucose showed no influence on it(P>0.05).3. Among all grou Ps, the damage levels of ileac mucosa increased significantly in a time-de Pendent manner(P<0.05), Positively correlated with the changes of D-lactin and DAO concentrations in Peri Pheral bloods sam Ples.4. The mitochondrial im Pair levels among different grou Ps were also significantly intensified in later time Points(P<0.05), consistent with the trends of D-lactin and DAO concentrations.Conclusions:1. Craniocerebral firearm injury can cause the terminal ileum mucosa damage, and with the extension of time and is aggravating, causing intestinal mucosal permeability increasing.2. High body tem Perature and humidity could aggravate the damage to teminal ileum. has promoting effect to intestinal permeability increase.However, high levels of blood glucose reveals little regulatory effects on the Permeability of terminal ileum mucosa.
