Objective: To investigate the proliferation in human bladder cancer cell line EJ induced by different concentrations of THP and to study the influence of THP on the expressions of Livin and Caspase-3 protein,and discuss the possible mechanism of action. Methods : EJ cells were cultured in vitro,and incubated with different concentrations of THP(0,0.1,1,10μg/ml),respectively;The morphological changes of the EJ cells was observed by inverted microscope;The cell proliferation was measured by growth curve;The distribution changes of EJ cells cycle was tested by flow cytometry;The expression of Livin and Caspase-3were detected by using reverse transcription-polymerase chain reaction(RT-PCR) assay, the expression of Livin and Caspase-3 was detected by Western Blot. Results: EJ cells proliferation has obvious inhibitoried by THP and presents the obvious time and concentration dependent manner(P < 0.05);Flow cytometry detection showed that THP could make the bladder cancer EJ cell block in G2/M phase; RT-PCR and Western Blot results all show that THP can significantly inhibit Livin gene expression and induction Caspase-3 geneexpression. Conclusions: The key mechanisms of Pirarubicin inhibit EJ cell proliferation and induce apoptosis may be related to downregulate the expression of Livin protein and to increase the expression of Caspase-3 protein of EJ cells, and then causing cell cycle stop in G2/M phase.
