The Effect Of Curcumin Combined With Pirarubicin On Bladder Cancer Cell In Vitro

Objective:The aim of this study is to investigate the effect of Pirarubicin combined with Curcumin on proliferation and apoptosis of bladder cancer cell lines,T24 cells、J82cells and 5637 cells in vitro,to explore its mechanism,provide a new solution for bladder cancer postoperative perfusion treatment. Methods:MTT assay was applied into detect the inhibitory growth rate of the cells,Cell apoptosis and the distribution of cell cycle was detected by flow cytometer using AnnexinV-FITC/PI detection kit. Results:Pirarubicin and Curcumin could respectively inhibit the growth of T24 cells、J82cells and 5637 cells and cause apoptosis significantly. The combination of Pirarubicin and Curcumin could inhibit the growth of bladder cells significantly more than the single effect.The results of flow cytometer show that the combination of Pirarubicin and Curcumin result in the cell apoptosis more than the sigle drug group(p<0.05).The combined application of Pirarubicin with Curcumin suggested that bladder cancer cells were arrested at S sphase or G2 sphase. Conclusion:1、The combination of Pirarubicin and Curcumin have synergistic effects on the proliferation,and the effects have time-effect and dose-effect relationship.2、The combination of Pirarubicin and Curcumin result in the cell apoptosis more than the sigle group,the rationality of the two medicine combined application was obvious.3、The mechanism of the two medicine combined application may connected with the affection the cell cycle.
