Background:Bladder cancer(cancer Bladder) is the most common malignant tumor in the urinary system. It is also one of the ten common tumors in the whole body. Its incidence rate is increasing year by year in China. Table for early type of bladder cancer, underwent routine after urinary tract electric cutting microscopically, the tumor resection(TURBT) get treatment of superficial bladder cancer, but the single operation, recurrence rate is high, the combination of postoperative intravesical instillation of pirarubicin and other rules of the bladder can reduce the recurrence probability, that part of pirarubicin combined therapy have a synergistic effect in treatment of patients with bladder cancer. VitK3 for human liver collagen synthesis enzyme must material, participate in related synthesis of coagulation factors, with the increase in diuresis detoxification function, involved in cell membrane material to repair function. The recent many scholars at home and abroad reports, vitamin K3 on certain tumor cells proliferation has inhibition, and induce cell apoptosis. In recent years, the newly discovered first with anti oncogene PTEN phosphatase activity, it is not only the characteristics of oncogene tumor suppressor, but also has phosphatase activity, plays an important role in the regulation of cell proliferation and death, cell migration and adhesion. Although some progress has been achieved in the treatment of bladder cancer, but after operation in the patients with high recurrence rate has become one of the major problem facing the bladder cancer clinical treatment. Therefore, exploring VitK3 combined with pirarubicin(THP) enhanced the inhibitory rate of tumor, reduce the complications related to the bladder, and explore the PTEN gene expression in pirarubicin and combined treatment of attack becomes the nowadays, it is an important aspect.Objective:Research VitK3 combined with pirarubicin for epithelial carcinoma BIU-87 cells and analysis with PTEN gene expression correlation and provide basis for clinical combined with bladder perfusion and bladder related gene expression and regulation.Methods: Culture of human bladder cancer BIU-87 cells. Using MTT assay Sai Zuolan were screening out of pirarubicin and vitamin K3 effect on BIU-87 cells after 3 days of the most appropriate drug concentration; blank control group was set, pirarubicin group, vitamin K3 group and combination group. MTT assay was used to test test each cell survival rate. Using flow cytometry observation of human bladder cancer BIU-87 cell line apoptosis of cultured. Cell cycle analysis by flow software; by Q-PCR analysis in each experimental group of human bladder cancer BIU-87 cell PTEN gene expression; through the expression of PTEN protein Western blot detection each experimental group of BIU-87 cells.Results : The drug concentration in 1mg/L pyran adriamycin concentration of 100mg/L can effectively inhibit the proliferation of BIU-87 cells(P<0.05); K3 drug concentration of Vitamin in the concentration range of 5 umol/L to 20 umol/L can effectively inhibit the proliferation of BIU-87 cells(P<0.05); Vitamin K3 combined with pirarubicin for better drugs inhibit BIU-87 cell proliferation effect(P<0.05) the concentration of 1mg/L; pirarubicin intervention can promote the apoptosis of BIU-87 cells(P<0.05), the cell cycle in the G2/M phase(P<0.05), the Vitamin K3 concentration of 5 umol/L dry pre BIU-87 cells can promote the apoptosis(P<0.05), but the cell cycle compared with the control group had no significant difference(P>0.05 Vitamin, K3) combined with pirarubicin intervention BIU-87 cells can make it more apoptosis(P<0.05), the BIU-87 fine Cell cycle arrest in G2 / M phase(P < 0.05); using Tetrahydropyranyl adriamycin 1mg / L concentration intervention BIU-87 cells can improve the expression levels of PTEN m RNA and protein(P < 0.05) and the vitamin K3 5 umol / L concentration intervention BIU-87 cells can improve the PTEN m RNA and protein expression levels(P < 0.05), vitamin K3 with pirarubicin intervention of adriamycin BIU-87 cells can more improve the expression levels of PTEN m RNA and protein(P < 0.05);Conclusion: A certain concentration of vitamin K3 and pirarubicin combined effects can significantly inhibit the proliferation of human bladder cancer BIU-87 cells, improve the PTEN expression prompted BIU-87 cell apoptosis and cell cycle arrest in G2 / M phase of the cell cycle.
