Bleeding is the most common phenomenon in surgery and emergency treatment. Rapid and effective hemostasis could reduce the chance of wound infection and was the key of saving patients’ life. So the efficiency and versatility of hemostatic could be very conductive to reduce death rate. Then the study of hemostatic materials got the high attention of medical field. Silk fibroin, which got excellent biocompatibility, was widely used as a kind of natural biomaterials. To study the potential of silk fibroin in hemostatic materials, this paper was devoted to study the preparation and properties of porous regenerated silk fibroin loaded with Ca2+.With silk fibroin crosslinked by PEG-DE at a certain ratio, we got porous silk fibroin materials by freezing-drying method. The flexibility of silk fibroin materials was improved by the adding of PEG-DE, which got flexible chain. Besides, porous structure endowed compressibility and water absorption with silk fibroin materials and provided space for the adhesion and aggregation of platelets. Materials with increased softness could reduce discomfort and pain; Materials with increased water absorption can absorb water in the blood quickly, increasing blood viscosity and promoting blood clotted; Materials with increased platelet adhesion space could aggregate a large number of platelet in a short time and activate coagulation factors, resulting rapid hemostasis. The different proportion of 6% of the silk fibroin solution Ca2+ preparation of porous materials in the 30 s release rate reached more than40%.Porous silk fibroin materials were fabricated with a certain mass ratio between silk fibroin and Ca Cl2(100:0;100:5;100:10;100:15;100:20). And then the water absorption, expansion, compressibility of materials, Rabbit ear wound bleeding test were studied. The results indicated that porous silk fibroin materials(silk fibroin concentration: 6%) had better water absorption and compressibility, but general expansion property. This paper studied the relationship between the content of Ca2+ and plasma clotting time and the weight of coagulum. The results showed that as the content of Ca2+ increased, clotting time decreased and the weight of coagulum got increased. Besides, porous materials(silk fibroin: Ca Cl2= 100:10,100:15) could make the plasma completely solidified, resulting rapid hemostasis.MTT assay suggested that materials(silk fibroin: Ca Cl2=100:0; 100:5; 100:10; 100:15) had no obvious cytotoxicity. Antibacterial experiments showed that porous silk fibroin materials loaded with Ca2+ has a certain inhibition effect on bacterial.
