Balanophora involucrata Hook. f. is a perennial parasitical succulent herbaceous plant which belongs to Balanophoraceae, Balanophora, it is a kind of Tujia medicine which including four medicines(Balanophora involucrata Hook, f., Diphylleia sinensis H.L.Li, Paris polyphylla, Trillium tschonoskii Maxim), its aliases are "wenwangyizhibi, qixinji". It is distributed in Jiangxi, Hubei, Hunan, Guangxi, Sichuan and Yunnan provinces. In Hubei the concentration distribution is in enshi and shennongjia region, it grows at an elevation of 1600-2000 m in cold and damp environment, it often parasites on the root of rhododendron and China berry. All parts of the plant can be used as a medicine, in the folk it is used in the treatment of traumatic hemorrhage, stomach pain, irregular menstruation, dysentery, and hemorrhoids, it also can be used as a tonic. Balanophora involucrata is widely used in Tujia folk, it has curative effect, is one of the traditional valuable Chinese herbal medicines. From recent years research, the major constituents of Balanophora involucrata has biological activities include anti-inflammatory and analgesic, sober-up and liver protection, anti-aging and anti-fatigue effect, antibacterial action and anti-asthmatic effect and so on.Objectives To study the chemical composition of Balanophora involucrata Hook. f. systematically and conduct preliminary study of the main composition on cytotoxicity.Methods The dried plant of Balanophora involucrata Hook. f. was percolated with 95% EtOH and portioned with petroleum ether, ethyl acetate, n-butyl alcohol fractionally. Constitunets were isolated and purified by thin-layer chromatography, silica gel column chromatography, PHPLC and recrystallization. Their structures were identified on the basis of their chemical and physical properties as well as the spectra data of MS, IR, UV, 1H-NMR, 13C-NMR et al.Results Seventeen compounds had been isolated from the plant of Balanophora involucrata Hook. f., they are:lupeol acetate (1), stearic acid (2), β-sitosterol (3), daucosterol (4), gallic acid (5), ethyl brevifolin carboxylate (6), methyl gallate (7), eriodictyol (8), trilobatin (9), calycosin-7-O-β-D-glucoside(10),3’,4’-dihydroxy flavonoids-7-O-β-D-glucoside (11),3-hydroxy-phloridzin (12), phloridzin(13), quercetin(14), genistin (15), luteolin-7-O-β-D-glucoside (16), (2R)-eriodictyol-5-O-β-D-glucopyranoside (17).Conclusion Seventeen compounds had been isolated from the plant of Balanophora involucrata Hook. f.. Compound 5 was isolated from Balanophora involucrata Hook. f. for the first time. Compound 2,6,10,11,15,16 were isolated from Balanophora for the first time. In addition, part of compounds isolated from the plant Balanophora involucrata Hook. f. were conducted a preliminary study of antitumor activity, this provides the evidence for the further utilization and research.
