| Euphorbiae Humifusae Herba(EHH),a commonly used and traditional Chinese medicine,is the dry whole plant of Euphorbia humifusa Willd.or Euphorbia maculata L.,which is included in Chinese Pharmacopoeia(Volume one,2015 edition).The related modern pharmacological research indicated that EHH had a variety of functions,such as antibacterial action,antiviral properties,inhibition and scavenging the oxygen free radicals.It has been reported that EHH contains many components such as tannin,methyl gallate,quercetin,glycosides and polysaccharide,of which the particular concern is the polysaccharide of EHH.It was a kind of important active substance to ensure its characteristic efficacies,since the curative effects of EHH are largely similar to some known activities of Chinese herb polysaccharides.However,little information published till now about the quantitative and separation optimization of EHH polysaccharide,let alone its bioactivity studies.So a deeper research on polysaccharide from EHH was very necessary for offering basic data to investigate the structure-activity relationship and the exploitation of the easily obtained herb resource.The paper systematically studied the extraction and degradation optimization of EHH polysaccharides,and its prebiotic activity on the regulation of intestinal balance was further explored.Chapter one: The textual research,chemical composition and pharmacological effects of EHH,and the activities and development of polysaccharide,oligosaccharide and monosaccharide from Chinese herbal medicines were summaried;the extraction optimizarion method of polysaccharide-Response Surface Methodology and the general degradation methods of polysaccharide were overviewed.Chapter two: The extraction process of EHH polysaccharide was optimized to improve the yield of polysaccharide.Based on the Plackett-Burman design(PB),the key factors were searched from the multivariables,and then Box-Behnken design(BBD)with three-factors and three-levels was used to further optimize the extraction conditions of EHH polysaccharide.Chapter three: The degradation conditions of polysaccharide were optimized to obtain different degrees of degradation products.Partial degradation and full degradation conditions were optimized by orthogonal test and the obtained product was initially qualitatively determined.Chapter four: Through the methodological verification,a capillary zone electrophoresis method was established which was simple,sensitive and efficient,and could be used for the analysis of the monosaccharide composition of the polysaccharide.Chapter five: The polysaccharide and its degradation products were evaluated to have a certain prebiotic activity by promoting helpful microbial populations and suppressing harmful bacterial growth assay.Chapter six: Conclusion and expectation.This paper could provide reference for the primary structure research and quality control of EHH polysaccharide.The results of the activity experiment partially revealed the mechanism of EHH in the treatment of diarrhea,and indicated that EHH polysaccharide was a kind of potential prebiotics,which could be further developed as a kind of green,functional additive. |
