Objective Observe the content and activity of colony-stimulating factor(G-CSF) and macrophagecolony-stimulating factor(M-CSF) in the CTX-induced mice through the different moxibustion time and moxibustion height. Research the dose effect relationship of moxibustion in animal experiments,to find out the best moxibustion time and moxibustion height, optimizated the dose effect standard of moxibustion, provide methodological basis for animal moxibustion experiment. Methods Fourty-two clean male Kunming mice, according to weight,randomly divided into six groups,A1B1 group(2cm、2min),A1B2 group(2cm、3min),A1B3 group(2cm、4min),A2B1 group(3cm、2min),A2B2 group(3cm、3min),A2B3 group(3cm、4min),seven of each group. According to dosage of 100mg/kg/d, abdominal injection of cyclophosphamide(According to the weight of mice,the concentration is 5mg/ml,for three days,after discontinuation of 4 hours on the third day,the model was sucessful,induced the bone marrow suppression model. Acupoints “Dazhui”( GV 14), “Geshu”( BL 17), “Shenshu”( BL 23) and “Zusanli”( ST 36) were used for treatment with moxibustion according to the group,A1B1 group the moxibustion height is 2cm,the moxibustion time is 2min;A1B2 group the moxibustion height is 2cm,the moxibustion time is 3min;A1B3 group the moxibustion height is 2cm,the moxibustion time is 4min;A2B1 group the moxibustion height is 3cm,the moxibustion time is 2min;A2B2 group the moxibustion height is 3cm,the moxibustion time is 3min;A2B3 group the moxibustion height is 3cm,the moxibustion time is 4min,and the daily treatment at the same time,each group treats 1 times a day, for 5 consecutive days.Each group draw blood from caudal veinto artificial detect white blood cells in peripheral blood,on the second day after five days of treatment,killed mice of each group,picked the eyeballs,got serum. ELISA method was uesd to detect the content of G-CSF and M-CSF in the serum.SPSS18.0 for Windows is adopted to analyze the data.Result1 After inject CTX, the number of peripheral blood leucocyte decrease in the six groups of mice compared with pre injection, and to minimize the number after chemotherapy, compared with before modeling, after making animal model, P values were less than 0.05,shows that the models success.2 Along with the moxibustion therapy,the number of peripheral blood white cells in the six groups of mice all showed increasing trend,after five days’ treatment the number reached the hightest,and more than the normal level before chemotherapy; but overall effect of moxibustion on the number of peripheral white blood cells in the CTX-induced mice,there were no different between moxibustion time and moxibustion height(the P value is greater than 0.05).3 After five days’ moxibustion treatment, the content of G-CSF increased in the serum of CTX-induced mice, and the best moxibustion time is 2min(P values were less than 0.05),but the moxibustion height was no significant difference(the P value is greater than 0.05).4 After five days’ moxibustion treatment, the content of M-CSF increased in the serum of CTX-induced mice, and the best parameter combination for moxibustion stimulation: the moxibustion time is 2cm(the P value is greater than 0.05), the moxibustion height is 3min(the P value is greater than 0.05).Conclusions1 Before making animal mode, the number of white blood cells were no significant difference between the six groups of mice, after modeling, the content of peripheral blood white cells were decreased in each group of mice, and to minimize the number after chemotherapy, shows that the chemotherapeutic agent cyclophosphamide could inhibit the hematopoietic,bone marrow’s function, destroy the hematopoietic cell,kill white blood cells.2 Moxibustion can regulate the colony stimulating factor G-CSF and M-CSF,increase thecontent of G-CSF and M-CSF in the surum,and G-CSF and M-CSF can stimulate the bonemorrow hematopoietic stem/progenitor cell proliferation,differention.then through themaoxibustion treatment,the result effectively improve the bone marrow suppression afterchemotherapy, recover the hematopoietic function,increase the number of peripheral bloodcells.Form the experimental point of view,there were no difference between moxibustion time and moxibustion height,it shows that the increasing of number of white blood cells in the CTX-induced mice associated with therapeutic effect of moxibustion height.3 On the effect of increasing the content of G-CSF in mice serum, between the moxibustion time and moxibustion time,there is no cross effect,the moxibustion time is 2min, the effect is the best,but there was no difference with the moxibustion height.4 On the effect of increasing of the content of G-CSF in the mice serum, between the moxibustion time and moxibustion time,there is cross effect,and the best moxibustion time is 3min,the moxibustion height is2 cm, the effect is the best.
