Purpose: to complex factors were used to produce cold coagulation stagnation ofphlegm and blood stasis combined with myocardial ischemia animal model, to observe theinfluence of Guanxin Fu Han Granule on myocardial injury in animal models, to investigatethe drug on myocardial injury induced by complex factors of the protective effect andmechanism.Method:60Wistar rats, were randomly divided into6groups: control group, modelgroup, positive control group (Yangxinshi tablets), Guan Xin Fu Han granule of high, mediumand low dose group,10rats in each group, the administration period, except the blank group,other5groups of animal fed with high fat diet,2freezer frozen hours daily, for6weeks, afterthe last administration, in addition to the normal control group, the rest groups animalsubcutaneous multi-point injection of pituitrin (10u/kg body weight), models of myocardialischemia. OUTCOME MEASURES:(1) related to the severity of the index ofelectrophysiological detection and myocardial injury: the model rats before and after thestandard lead II ECG as observation indexes, calculation of ECG ST segment deviation oftotal millivolt values (∑-ST) said the extent of myocardial injury. The pathologicalexamination: heart, aorta, formaldehyde fixation, HE staining to observe the myocardial fiberfracture, edema, under the light microscope, severe myocardial fiber solubility necrosis.Observation of myocardial ischemic area and the degree of damage under the microscope, themyocardial damage degree is determined by the integral method. The blood biochemistrydetection: detection of serum myocardial enzyme (LDH, CK, AST), myocardial injury markerprotein, troponin: troponin I (cTnI), ischemia modified albumin (IMA) etc.. The lightmicroscope to observe the morphological changes of myocardial injury;(2) associated withthe protective mechanism of myocardial ischemia injury index: the antioxidant effect ofanimal: detection of serum GSH-Px, ELISA SOD, MDA enzyme change; detection of antiapoptosis: the expression of apoptosis related genes Bcl-2, Bax protein; homocysteineproteinase3.Result: Guanxin Fu Han granule(GFG) can reduce the sum of animal model of STsegment elevation millivolt values (∑-ST). To reduce the content of serum myocardial enzyme. Reducing the content of myocardial injury markers. Histological observation showedthat myocardial injury score lower than those in the model group. Prompt Guanxin Fu Hangranule (GFG) orally can lessen the degree of myocardial injury in animal modelsConclusion: Guanxin Fu Han granule(GFG) can lessen the degree of myocardial injuryin animal models, and has a protective effect on cold coagulation of phlegm and blood stasissyndrome complicated with myocardial ischemia animal myocardial cell injury. Deathmechanism of Guanxin Fu Han Granule on the protective effect of myocardial cell damageand the adjustment of the oxidase activity, inhibition of cell injury, repair of myocardial injuryfactors DNA and regulation of the platelet activating factor activity of regulating blood.
