Background and objectiveCoal workers pneumoconiosis is a kind of pneumoconiosis, which can cause long-term chronic and irreversible lung fibrosis.Many studies have been done for the development and progression of pulmonary fibrosis pneumoconiosis, but the exact mechanism is still not very clear. In recent years, a large number of studies hadve shown that the process of pneumoconiosis was closely related to the occurrence and changing in immune function.With the development of the immune technology, phage antibody library technology becomes a mature technology.Early application of the research group of this project successfully built a human phage antibody library of coal workers pneumoconiosis (CWP), and screened six human strongly positive monoclonal antibody, one of them wasGranzyme B. GzmB are a kind of exogenous serine protease, which come from the cytoplasmic GzmAnules of Cytotoxic lym-phocytes (CTLs) and natural killer cells (NK).Gilani found that the expression of GzmB concentration was increased significantly in fibrosis of lung tissue.We decided to study the levels of GzmB in serum of CWP and workers exposed to dust, to explore the significance in the pathogenesis of pneumoconiosis.Methods1. Respondent:All of230pneumoconiosis observed people were from2cities of Henan province.328coal workers’ pneumoconiosiscases were all from the same area.309exposed workers were from the same area who had dust exposed history.393healthy subjects as control group, who were from onehospital of Henan province.2. All subjects conducted a questionnaire survey, including:personal general situa-tion, dust operating history, diagnosis, medical history and family genetic history. Serum samples collected:All subjects collected peripheral venous blood, sepa-rated and collected the serum. Then placed the serum in-80℃refrigerator.3. Detect the expression levels of GzmB in all subjects serum by ELISA 4. Statistical methods:SPSS21.0statistical software for experimental data entry and statistical analysis, and the significant level was0.05.Results1. The CWP group GzmBOD value are higher than exposed group and control group, the differences were statistically significant (P<0.05).2. In the CWP group, age20~group and age40-group GzmBOD value are both lower than age60-group, the differences were statistically significant (P<0.05).3. The expressions of GzmB in different work group were undifferentiated.4. The influence of different work group, dust expoed time, smoking and drinking on the expression of GzmB and its antibody:In CWP group, dust expoed time and have varying degrees of impact in GzmB OD value.ConclusionThe serum levels of GzmB in CWPs and exposure group are increased. The ex-pression of GzmBin serum of CWPs are positive correlated. Serum levels of GzmB may be related to dust exposure et al.
