Objective To investigate the Gualou Xiebai Banxia Decoction (GXBD) effect on myocardial ischemia reperfusion injury in rats induced angiogenesis factor VEGF, FGFMethods50male SD rats were randomly divided into five group, A group:sham operation group, B group:model group, C group:ischemic preconditioning group, D group:pretreatment group, GXBD low dose, E group:pretreatment group and high dose GXBD,8rats in each group (death removed). Every day gavage for1times, continuous7d,Group A, group B, group C to equal volume of physiological saline, D group4.6g·kg-1dose of Gualou Xiebai Banxia soup decoction, E group18.4g·kg-1.After the last administration of1H for coronary artery ligation in rats anterior descending30min, reperfusion90min production of myocardial ischemia reperfusion injury model. Observation of ST segment of ECG changes the rats in the surgery; after modeling, the rats were measured serum lactic dehydrogenase (LDH), bradykinin (BK) levels; to observe the changes of myocardial pathomorphology of rats; determination of myocardial tissue of rats in each group of micro vascular density (MVD); to observe the expression of VEGF in myocardial tissue of rats in each group, FGF protein; to observe the myocardial tissue of rats in the groups VEGF, FGF gene expression.Results1. Compared with A group, B group each time of ST segment elevation (P<0.01), serum LDH level was significantly increased (P<0.01), BK (P<0.05) level was significantly decreased, the myocardial cells showed typical pathological changes after staining of HE, MVD increased (P<0.01),VEGF, FGF protein and gene expression increased (P<0.05, P<0.01). 2. Compared with B group, C group and D E group, each time the ST segment (P<0.05), serum LDH levels were significantly lower (P<0.01), BK level increased significantly (P<0.01), HE damage degree of myocardial cells was significantly reduced after staining, MVD increased (P<0.05, P<0.01), significantly increased the expression of VEGF FGF, protein and gene (P<0.05, P<0.01).3. Compared with C group, D group while the ST segment (P<0.05), the level of LDH was significantly decreased (P<0.01), HE damage degree of myocardial cells was significantly reduced after staining, MVD increased significantly (P<0.01), significantly increased the expression of VEGF, FGF protein and gene (P<0.01); E group and C group compared the level of BK were significantly increased (P<0.01), significantly increased the expression of FGF gene (P<0.01), more than the difference was not statistically significant.Conclusions GXBD can promote the expression of VEGF, FGF protein and gene, promote angiogenesis, pathological changes of myocardial cells, reduce the content of LDH, increase the content of BK, on ischemia-reperfusion myocardium protection effect, and the low dose was better than high dose.
