Objective To study the influence of Jishang Particle on the expression of Caspase-3、TGF-β1and MMP-9and the motor function after spinal cord injury in rats. To explore the effect and mechanism of Caspase-3、TGF-β1and MMP-9in Secondary injury after spinal cord injury And the affect of the traditional Jishang Particle in the treatment of spinal cord injury.Methods100healthy adult SD rats were randomly selected24rats as the control group, the rest of the rats using modified Allen’S blow machine manufacture SCI rats model, building a successful72rats were randomly divided into model group, methyl prednisolone group and Jishang Particle group. Respectively were given to control group, model group, normal saline lavage, methyl prednisolone group tail intravenous drugs, Jishang Particle particles which drug lavage. In experiment1,3,7,14d after using four different time points of BBB behavioral functions; Another batch to be put to death in the rat, remove SCI with HE staining to observe the injury of spinal cord tissue pathological changes, detected using immunity texture chemistry methods staining Caspase-3, TGF-β1positive cells expression, enzyme-linked immunosorbent experiment ELISA (clip art) to detect the content of MMP-9, the apoptotic cells in situ end labeling method of marking. Application IPP6.0Caspase-3groups in terms of pathological image analysis software to determine the Integral optical density value of positive product IOD (Integral optical density) and TGF-β1positive product density optical density value (OD value), and then averaging as the sample IOD values, OD value.Results (1) the BBB score results:after SCI, compared with control group, model group, Jishang particles, methyl prednisolone group BBB score was significantly lower (P<0.01); Compared with model group, Jishang particles, methyl prednisolone group score higher (P<0.05); Compared with methyl prednisolone group, SCI group after1,3d Jishang particles low scores (P<0.05);7,14d after SCI, Jishang score high particle group (P<0.05).(2) in terms of average IOD values and cell apoptosis rate results:after SCI, numerical on the high side compared with control group, model group (P<0.01), methyl prednisolone and Jishang group average value is on the high side (P<0.05); Compared with model group, methyl prednisolone and Jishang group average value is low (P<0.05); Compared with methyl prednisolone group, SCI group after1,3d Jishang particles average values on the high side (P<0.05),7d numerical approach (P>0.05), while14d, Jishang particle group average value is low (P<0.05).(3) in the mean OD value results:after SCI, compared with control group, model group, the Jishang particle group, methylprednisolone group average OD value (P<0.05); Compared with model group, Jishang particle average OD value higher (P<0.05), methylprednisolone group compared with the average OD value close (P>0.05); Compared with methylprednisolone group, Jishang particle average OD value higher (P<0.05).(4) the content of MMP-9results:after SCI, compared with control group, model group, the Jishang particle group, MMP-9levels of methyl prednisolone group were significantly higher (p<0.01); Compared with model group, Jishang particles, methyl prednisolone group of MMP-9low levels (P<0.05); Compared with methyl prednisolone group,3d,7d after SCI Jishang particle group of MMP-9levels close to, there was no statistically significant difference (p>0.05).14d after SCI, Jishang particle group of low content of MMP-9(P<0.05).Concludes1. Jishang Particle can restore function of behavior of rats after SCI, exact effect, is a kind of effective treatment of spinal cord injury (SCI) granule of Chinese medicine preparations.2. The model rats after spinal Jishang Particle can inhibit the SCI Caspase-3, the expression of MMP-9, reduced the amount of harmful factors, spinal cord injury on the expression of TGF-β1has a promoting effect, increase the number of the protection factors, both supplement each other effectively alleviate the SSCI in general; 3. The Jishang Particle can obviously reduce the expression of apoptosis cells and promote spinalcord function recovery;4. Jishang Particle and methylprednisolone can after SCI by Caspase-3, TGF-β1and expression of MMP-9and reduce the influence of SSCI. Spinal injury pellet in rats with SCI early treatment effect is not as good as methylprednisolone, later can more effectively prevent the further development of the secondary SCI, part of the recovery of motor function, promote the rats for the complementary advantages, traditional Chinese medicine and western medicine SCI provides experimental basis.
