Abstract:Objective:In order to observe the immune function changes rule in lung caner patients,the study has investigated their peripheral lymphocyte subsets proportion and absolute counts, with other clinical data such as pathological classification, staging and blood routine examination etc, helps to diagnose earlier and seek new therapy methods. Methods:twenty-two patients with primary lung cancer pathologically or cytologically confirmed for the first time and twenty healthy controls at the same period were enrolled from April2013to October2014,we divided them into two groups and everyone were collected in the morning on an empty stomach blood, anticoagulation. flow cytometry was used to measure the percentage and number of lymphocyte subgroups(CD3+、CD3+CD4+、CD3+CD8+、CD4+/CD8+、NK cell and B cell), and full automatic blood cell analyzer was used to determine the count of WBC、NEU、LYM in peripheral blood.Compare the expression of lymphocyte subsets in lung patients with different gender (male/female), age (>=65,<65) pathological type (SCCL/NSCCL) and staging (early/late).As to the patients who only accepted chemotherapy, we also collected samples of peripheral blood at different time point (pre-chemotherapy,4-7daysafter chemotherapy,21days after chemotherapy). All of the data were analyzed by SPSS17statistical software.Results:,1、Compared with the normal people, the percentage of CD3+, CD3+CD8+lymphocyte subsets in peripheral blood with lung cancer patients was significantly higher,(t=1.446,3.524, P0.015,0.001),the proportion of NK cells was significantly reduced (t=-3.320, P=0.002).No obvious differences were observed in CD3+CD4+、CD19+and CD4+/CD8+ratio(t value were2.542、1.109and-1.123, P>0.05).2、The level of CD3+、CD3+CD4+、CD4+CD8+、NK cell、B cell numbers in lung caner group were significantly lower compared with control group.(t values are-4.139、-4.154、-2.686、-6.607、-3.426、P<0.01)3、 Comparison among different gender、smoking history and pathological pattern has no obvious differences (P>0.05).The old group (aged>=65) patients with lung cancer expressed obviously lower CD3+CD3+CD4+, CD19+,CD4+/CD8+lymphocyte count than that of middle age groupage<65)(P<0.01).4、NK cell and B cell numbers decreased significantly in patients with early stage(P<0.05),CD3+、CD3+CD4+、CD4+CD8+count has no obvious alteration in early stage,however immune function declined further as the disease progress, and all the lymphocyte subsets count decreased significantly in the advanced stage (P <0.05).5、Neutrophil count in peripheral blood increased obviously compared with healthy control group while lymphocyte count declined obviously (t value were3.526、-6.132, P<0.01), and the WBC was higher than healthy control group, but no obvious difference was seen.6、CD3+、CD3+CD4+absolute numbers in lung cancer group increased obviously on the4th to7thday post-chemotherapy,(t value were-2.436、-3.178, P<0.05), NK cell decreased after therapy, however the count has no significance. All index come to the normal level on the21th day post-chemotherapy, and the expression of CD3+、CD3+CD4+in different point of time after chemotherapy has obvious difference (F value were4.714、6.372; P<0.05). Conclusion:1Lung cancer patients has a bad immune funcion, presenting with the number of all the lymphocyte subsets decreased.2、The level of CD3+、CD3+CD4+、CD4+CD8+with lung cancer patients decreased obviously in advanvced stage, revealing a bad immune function.3、Expression of lymphocyte subsets on different time point shows that CD3+、CD3+CD4+increased obviously on the early stage after chemotherapy and then come to the normal level on the21th day (P<0.05).Observation of immune function changes rule in lung caner patients after chemotherapy has important significance in diagnosis and treatment for clinical.
