Objective:To explore the protein expression of vascular endothelial growth factor (VEGF)in thelung adenocarcinoma cells (LAC)by the effect of an non-selectiveβ-blocker-propranolol(P).To reveal the effect of the non-selective β-blockers inhibiting the mechanisms of lungcancer angiogenesis,in order to provide with a theory and experimental basis in anti-tumorangiogenic therapy by the propranolol.Methods:Choose the human lung adenocarcinoma A549cells which are in logarithmic phase ofgrowth and grow in well addition,seed these cells into the6-well cell culture plate andcultured,during the period,change culture medium on time. Using the randomizationprinciple,we divide these cells into the experiment group and the empty control group.Under the inverted microscope,you can observe the growth state of these cells,when thenumber of adherent cells increase more than80%,suck out the cell culture from the6-wellplates,a volume of the different concentrations in propranolol(1,2,4,8,16ug/ml)which wasadded into each hole of the experimental group is2ml,and the hole of the empty controlgroup was added an equal volume of culture medium (the concentration of propranolol is0ug/ml),put these cells into the cells’incubator and brought them up. After these two groupswere brought up for24hours,you could disclose the protein expression of VEGF in thelung adenocarcinoma A549cells by way of immunohistochemistry’s method,observed itsconsequent in the experimental group and the empty control group between the propranololunused and propranolol used.Results:(1) In the experimental group and the empty control group,the lung adenocarcinomaA549cells present the protein expression of VEGF,its protein positive signals were mainly seen in the nuclear membrane surrounding the nucleus and cytoplasm.(2) Compared with the empty control group,after the treatment by way of thepropranolol,the protein expression of VEGF in lung adenocarcinoma A549cells of theexperimental group are reduced,its protein positive signal is reduced,but its expression donot alter the position.Conclusions:(1) After the normal lung adenocarcinoma A549cells are inoculated,their logarithmicgrowth phase can maintain about3-5days,and then,these cells begin to enter into thegrowth,at last,the growth rate of A549cells will be decreased. If we give the drug tointervene these cells,the growth rate of them will be decreased significantly, and they willenter into apoptotic state.(2) Generally,the VEGF receptor exists in vascular endothelial cell membrane of thelung adenocarcinoma A549cells,so VEGF can activate the pathway of the ERK/MAPK,inthis way,it can promote angiogenesis.(3) The positive cells and the more strongly positive cells which can possess theprotein expression of VEGF can be killed by the Propranolol,and then,the Propranolol canreduce the protein expression of VEGF in the lung adenocarcinoma A549cells.(4) With increasing concentration of the propranolol,the protein expression of VEGFin the lung adenocarcinoma A549cells will be reduced in a concentration-dependent.(5) The propranolol play its role in reducing the protein expression of VEGF,in thisway,the propranolol can restrain the angiogenesis,in the further step,it can inhibit theproliferation、growth and metastasis of the lung adenocarcinoma cells.(6) By the guidance of clinical application,some targeted drugs which are used in theclinical treatment can provide an important reference to us. Considering the application ofsome inhibitors in the clinical treatment,the role of VEGF in cells signaling pathway canbe interfered and blocked specifically,and then,it can provide some new ideas to theclinical treatment of the Lung Cancer.
