Objectives:To study the pathogenesis of osteoporosis using needling acupoint Guanyuan (CV4) on ovariectomized rats (OVX) by electro-acupuncture therapy, via the viewpoints of Estradiol impact on bone metabolism. The experiment also provide data prevention and treatment of postmenopausal osteoporosis for clinical purpose.Methods:24female SD rats aged4.5months,take out6rats randomly as the sham group.the left18rats after ovariectomy were divided into electro-acupuncture group, model group and sham group randomly,with6rats in each group respectively. Three months after ovariectomy and feeding, confirms whether ovariectomized rats with osteoporosis model were established. Treatment cycle of needling acupoint Guanyuan (CV4) on ovariectomized rats by electro-acupuncture therapy:once per day,20minutes each time and10times per treatment on Guanyuan group and non-acupoints group. There are3treatments in total with1day rest between each treatment cycle. Similar fat tissue weight around ovary with ovary weight of the OVX was removed from the sham group rats. Both groups were not given any treatment. After completion of this experiment, use injection draw blood from abdominal aorta.the blood centrifugal15minutes at3000r/minute,take the supernatant fluid,store in refrigerator at negative20degrees for inspection, use enzyme labeling immunosorbent assay method detecting E2、 ALP、 DPD individually.Results:1. The level of bone Biomechanical was detected:The level of bone Biomechanical between electro-acupuncture Guanyuan group and model group, it has significant difference statistically (P<0.05). The level of bone Biomechanical between electro-acupuncture Guanyuan group and shanm group,it has no significant difference statistically (P>0.05).The level of bone Biomechanical between electro-acupuncture Guanyuan group and electro-acupuncture Non-acupoint group, it has significantly difference statistically (P<0.05). The level of bone Biomechanical between sham group and model group,it has significant difference statistically (P<0.05).The level of bone Biomechanical between model group and electro-acupuncture Non-acupoint group, it has no significant difference statistically (P>0.05). The level of bone Biomechanical between sham group and electro-acupuncture Non-acupoint group, it has significantly difference statistically (P<0.05).2. The level of Serum-E2was detected:Compared with the model group, the level of Serum E2in electro-acupuncture Guanyuan group, it has significantly difference statistically (P<0.05). The level of Serum E2between electro-acupuncture Guanyuan group and sham group, it has no significantly difference statistically (P>0.05). The level of Serum E2between electro-acupuncture Guanyuan group and electro-acupuncture Non-acupoint group, it has significantly difference statistically (P<0.05). The level of Serum E2in sham group compare with model group, it has significantly difference statistically (P<0.05). The level of Serum E2between electro-acupuncture Non-acupoint group and model group, it has no significantly difference statistically (P>0.05). The level of Serum E2between sham group and electro-acupuncture Non-acupoint group, it has significantly difference statistically (P<0.05).3. The level of Serum-ALP was detected:Compared with the electro-acupuncture Guanyuan group, the level of Serum-ALP of model group, it has significant difference statistically (P<0.05). compare with electro-acupuncture Guanyuan group, The level of Serum-ALP of sham group, it has significant difference statistically (P<0.05). compare with electro-acupuncture Guanyuan group,the level of serum-ALP of electro-acupuncture Non-acupoint group, it has significant difference statistically (P<0.05). The level of Serum ALP in sham group compare with model group, it has no significant difference statistically(P>0.05). The level of Serum-ALP between model group and electro-acupuncture Non-acupoint group, it has no significant difference statistically (P>0.05). The level of Serum-ALP between sham group and electro-acupuncture Non-acupoint group, it has significantly difference statistically (P<0.05).4. The level of Serum-DPD was detected:The level of Serum DPD of model group compared with the electro-acupuncture Guanyuan group,, it has significant difference statistically (P<0.05). The level of Serum ALP of sham group compare with electro-acupuncture Guanyuan group,it has no significant difference statistically (P>0.05). compare with electro-acupuncture Guanyuan group,the level of serum ALP of electro-acupuncture Non-acupoint group, it has significantly difference statistically (P<0.05). Compared with the model group, the level of Serum DPD of sham group,it has significantly difference statistically (P<0.05). The level of Serum-DPD between model group and electro-acupuncture Non-acupoint group, it has no significant difference statistically (P>0.05). The level of Serum-DPD between sham group and electro-acupuncture Non-acupoint group, it has significantly difference statistically (P<0.05).Conclusion:1. electro-acupuncture Guanyuan can improve the bone biomechanical of model rats.2. electro-acupuncture Guanyuan can improve the serum-E2of model rats, so as to improve the bone lose sympotom of model group rats.3. electro-acupuncture Guanyuan can promote the serum-ALP level of model rats, so as to improve bone form.4. electro-acupuncture Guanyuan can compress the serum-DPD level of model rats,in order to decrease the bone absorb.5. compare to electro-acupuncture Non-acupoint group electro-acupuncture Guanyuan group can improve the serum-E2of model rats, so as to improve the bone lose sympotom of model group rats.
