Objective: To investigate the anti-irradiation effect and mechanism of asiaticoside invitro and in vivo.Methods:①NIH3T3cells were cultured. Before the radiation, the cells were treatedwith AS in different concentration respectively. The irradiation damage models of cellswere established by60Co gamma ray (15.0Gy, at a dose rate of2.0Gy/min), then theeffect of AS on those cells were determined by MTT assay. Total SOD activity detectionkit (NBT method) was used to detect the level of SOD in the cells lysate after the radiation.By flow cytometry, cell cycle kit was used to detect the effect on cell cycle of AS withdifferent concentrations after radiation.②Irradiation damage models of mice wereestablished by X ray (10.0Gy, at a dose of1.67Gy/min) in vivo. The protective effect ofAS against damage of irradiation were observed through the weight, the spleen and thymusindexes of mice; Splenocyte proliferation was determined by MTT assay; Intestinalepithelial tissue structures of mice were observed through hematoxylin and eosin (HE)staining at different time points (the fifth hour, the second day and the eighth day) afterirradiation.Results:①MTT assay showed that three different concentrations of AS (7.5μg/ml,15μg/ml,30μg/ml) had a protective effect on NIH3T3cells after irradiation. Threedifferent concentrations of AS (30μg/ml,60μg/ml,120μg/ml)could significantlyincrease the level of SOD in the cell lysate. Cell Cycles at48hours after irradiation, G1phase in the model group compared with normal group was significantly decreased, andthe proportion of S phase cells was elevated, namely S phase arrest, AS (15μg/ml,30μg/ml) could significantly improve the S phase resistance stagnation and enter the G2phase.②On the eighth day after irradiation, AS failed to stop the decline in bodyweight,but AS could increase the mice spleen index and thymus index. AS of large dosegroup (25mg/kg) could improve the spleen lymphocyte proliferation activity. On the morphological observation, AS could recover the intestinal epithelium organizationalstructure after irradiation at different time points (the fifth hour, the second day and theeighth day).Conclusion: AS has a certain anti-irradiation effect. The mechanism may be relatedto anti-oxidation,enhancement of immune function and improvement of cell cycledisorders.
