Objective:First of all, comparing the quality of different manufacturers Poria, Guizhi,Atractylodes, Licorice Granules; Secondly, to establish a content determination method ofLingguizhugan traditional soups and compound granules, formula granules, and comparingthe differences of chemical composition of the three kinds of preparations. And to furtherexplore the mechanism of action of anti-heart failure efficacy of the three kinds ofpreparations. Which provide some experimental basis for clinical application ofLingguizhugan compound granules and formula granules.Methods:1. By Appearance, soluble and labeling specifications, thin layer chromatography,determination, fingerprints and other aspects, the comprehensive comparative study wasdone on quality of Poria, Guizhi, Atractylodes, Licorice dispensing granule from differentplants.2. Using the strategy of boiling to prepare clinic traditional decoction, optimizing thetimes of boiling, the methods of enrichment and drying. Drawing up the technologicalprocess of preparing traditional decocton, compound granules. And simulating the strategyof the industrial production of dispensing granule decoction and combining with the modeof condensing and baking that had been sieved out to prepare the dispensing granuledecoction.3. HPLC was used to compare the difference of chemical composition andcharacteristic chromatogram of the three kinds of preparations, and the similarityevaluation of the three kinds of preparations were analyzed by the similarity software. Secondly, studying the experiment of detaching the formula,thus to explore the singlemedicinal material,s effect on fingerprints and to perform standard atlas source attribution.4. With70SD rats were randomly divided into normal control group, model group,heart failure, digoxin group, Lingguizhugan traditional decoction group, compound granulegroup, Granules group. In addition to the normal control group adopts tail vein injection ofADR which can cause heart failure. Each group was given the appropriate medication, after,the levels of BNP, Ca2+, BK, SOD, MDA were detected and the myocardial tissue wereobserved by light microscopy.Results:1. There were great differences from appearance, soluble and labeling specifications,thin layer chromatography, determination, and characteristic chromatogram betweendifferent plants.2. In HPLC, in three kinds of compositions, the content of cinnamic acid, licoriceglycosides, ammonium glycyrrhetate in dispensing granule are the highest. In thedetermination of characteristic chromatogram, the correlation coefficient of HPLCchromatograms between them was0.576,0.365. The main chemical substances of thetraditional decoction, compound granules and formula granule were similar but the contentwas different.3. The method has better precision and repeatability, Lingguizhugan soup traditionaldecoction, compound granules and granule formulations were identified34common peaks;similarity compound granule decoction of0.876and traditional recipe granules similaritywith traditional decoction of0.465on the characteristics of the three peaks and the numberof little difference, but chromatographic peak area, peak height were quite different.4. Compared with the normal control group, the rats’ serum of the heart failure modelgroup has elevated levels of BNP, BK, and MDA; and has lower levels of SOD, Ca2+;compared with model group, the other group can significantly reduce the level of BNP, BK,and MDA of heart failure model, and increase the level of SOD, Ca2+. And lightmicroscopy showed that the myocardial damage degree of others groups were significantlylighter than heart failure.Conclusion:1. Granules among enterprises with the uneven quality varieties, establish uniform quality standards is imperative.2. Lingguizhugan soup traditional decoction and its compound, index componentscinnamic acid, ammonium glycyrrhizin and licorice highest content in the recipeformulation granules granules, traditional soups and Compound Granule content less.3. Lingguizhugan soup traditional decoction and its compound granules is similar,there are some differences with the formula granules, consisting of the three main chemicalcomposition is basically the same, but there were large differences in ingredients.4. Lingguizhugan soup traditional decoction and its compound, formulation granuleswere able to improve the general condition of the body weight of rats with heart failure,mental, hair, etc., to enhance the body’s oxygen radical scavenging capacity, improvemyocardial blood supply, reducing myocardial oxygen capacity, reduce symptoms of heartfailure and other aspects of the effect is obvious. Generally comprehensive experimentalobservations in rats and test results of the indicators, with the traditional decoction andcompound granules effect is more obvious.
