Objective:By observing the changes of the expression of NPY and CGRP in rats hippocampus with depression after focal cerebral ischemia injury, to explore the mechanism of PSD treated by modified BYHWD,provide a experimental basis for the clinical treatment of PSD by Chinese medicine.Methods:SD rats were selected.Each rat’s weight was from200-240g.They were randomly divided into six groups:sham operated group,MCAO group,PSD group,Buyang Huanwu Decoction(BYHWD) group,BYHWD modified group and fluoxetine group.Made PSD model by useing middle cerebral artery occlusion(MCAO) method combining with Chronic unpredictable mild stress(CUMS) and solitary raise.To treat these rats with different medicine,and all the rats will be test the weight and sugar consumptions at7,14,21days point after operation.All rats were killed in22day after operation.The expressions of NPY and CGRP in rats hippocampus were measured by immunohistochemical staining.Results:1.All the rats with left middle cerebral artery occlusion,in varying degrees of right arm adduction, buckling and with right-circling objective signs, showed that the MCAO models succeeded.After combining with CUMS,rats showed the phenomenon of irritability, decreased activity, diet reduce gradually.2.PSD rats both weight and sugar consumptions were decreased,shows the typical symptoms of depression after stroke.After treat by BYHWD modified,BYHWD and Fluoxetine,they were all increased,BYHWD modified group is more obvious.Compared with the PSD group these groups increased significantly,there is a significant differences(P<0.01). 3.Immunohistochemical statistical analysis results showedrcompared with the sham operation group,the average optical density of NPY in PSD hippocampus decreased greatly while the average optical density of CGRP increased obviously(P<0.01).This implyed that the contents of NPY and CGRP in rats’hippocampus area had abnormality variation after PSD.After treated by modified BYHWD,the average optical density of NPY increased greatly while the average optical density of CGRP decresed obviousIy(P<0.01).Conclusions:1.Modified BYHWD can improve the depressive symptoms of the rats with depression after focal cerebral ischemia injury,increase the weight of rats,improve the reactivity of reward of the rats.2.Modified BYHWD can enhance the expression of NPY,inhibit the expression of CGRP in the rats hippocampus with depression after focal cerebral ischemia injury.To improve the depressive symptoms of the rats by control the release of cerebral vessels neuropeptide.
