| In this study, we screen known and pr oven disease-related genes using online Mendelian Inheritance in Man database(OMIM), simultaneously apply cytoscape software to create a protein-protein interaction networks based on biological function, detect molecule complex may be includ ed in th e network.The relevant molecular com plex biological pathways that m ay be involved we re predicted that can provide a basis for further explain the mechanism of chronic myeloid leukemia development.Genes involved in chronic m yelocytic leukemia were selected from OMIM database. Literature mining for the genes was perfor med by Agilent Literatu re Search plugin. Protein-protein interaction network of chr onic myelocytic leukemia was established by Cytoscape. The molecular complexes in the network were detected by Clusterviz plugin and the pathways enrichment of molecular complexes were performed by DAVID online.To study the key pathway for JAK2-ST AT3, we use MTT, streaming,weston-blot methods in vitro experiments to observe the effect of rhubarb Zhechong pill on K562 cell proliferation and expression of JAK2, STAT5. The results showed that CML is a variety of signaling pathways, ne twork system consisting of multiple gene regu lation synergistic complex process, not simply a gene or a particular signaling pathways control.In the signal network, JAK2- STAT5 is likely to be " a key regulatory point", and rhubarb Zhechong pill can suppress the value of K562 cells, the m echanism may be related to its in herent impact on JAK2- STAT5 expression.Our study have shown that chronic myeloid leukemia has complex genenetworks,and JAK2- STAT5 is a key node.One of the inhibition mechanism Zhechong rhubarb pill on K562 cell proliferation is by reducing expression of JAK2-STAT5,which can provide ideas for further study. |
