In this study, we construct the gene/protein interaction networks and analyze the molecular complexes, which are included in the network, for digging the relevant molecular signaling pathways. Intervene the key signaling pathways by rhubarb Zhechong pill to explore the mechanism of which to treat primary myelofibrosis.We screen known and proven disease-related genes using Online Mendelian Inheritance in Man database(OMIM). Literature mining for the genes was performed by Agilent Literature Search plugin. Gene/protein interaction networks of primary myelofibrosis were established by Cytoscape. The molecular complexes in the network were detected by Clusterviz plugin and the pathways enrichment of molecular complexes were performed by DAVID online. We use rhubarb Zhechong pill to intervene JAK-STAT signaling pathway, which were remaindered by molecular complexes. Before and after treatment, apply TUNEL method to detect the apoptosis of bone marrow mononuclear cells and use ELISA and Western blot method to observe the effect of rhubarb Zhechong pill to the expression of STAT5 protein.Molecular complexes are involved in KEGG pathways that are related to JAK-STAT signaling pathway, cytokine regulation, immune function regulation and et al. The result of study indicates that the apoptosis rate of bone marrow mononuclear cells goes up and the expression quantity of STAT5 protein decline after using rhubarb Zhechong pill for primary myelofibrosis patients.The construction of gene/protein interaction networks for complex disease can provide reliable basis for clinical and experimental research. Rhubarb Zhechong pill can make a positive effect for primary myelofibrosis patients by changing JAK/STAT signaling pathway biology, which may be one of the mechanisms about its anti-tumor effect.
