YiQiJianPi Formula is a fundamental prescription of the study on optimization of treating chronic atrophic gastritis (CAG) with Traditional Chinese Medicine (TCM), which is from the 1 lth Five-Year Scientific Support Plan. It had efficacy of replen-ishing qi to invigorate the spleen and activating blood circulation. And it had shown satisfactory effect in treating CAG. Based on pre-clinical efficacy of this prescription, with its object of study, from theory and animal experiments, to prove its efficacy and provide material of experiments.Part I:Theoretical Study. First, we summarized from pathogenesis, diagnosis method, treatment principles and the choice of drugs about CAG in modern medicine. After analysis, we think it has obtained some achievements on pathogenesis and treatment of CAG in modern medicine. But it is still lack of specific methods in treating CAG. And then we summarized from pathogenesis, diagnosis method, treatment principles and the choice of drugs about CAG in TCM. More and more studies show specific advantages of TCM in CAG treatment. From the pathogenesis opinion of TCM, asthenia of the spleen and stomach is the basic pathogenesis of CAG, qi stagnation and damp-heat are important in the occurrence and development of CAG. The syndrome differentiation and treatment is the characteristics of TCM. YiQiJianPi is the basic treatment of CAG. Meanwhile, we analyze monarch structure and the pharmacological effects of TCM in treating CAG, it will provides a theoretical basis for the efficacy of YiQiJianPi Formula.Part II:Experimental Study Objective:To found rats model by MNNG with other factors and observe the effect of YiQiJianPi Formula on the RegI, PCNA and pathology, explore the possible mechanism for its efficacy. Methods:There were 60 raats divided into 6 groups. The treatment included modern medicine (folic acid 3.16mg/kg/d and teprenone 15.8mg/kg/d), the low dose of YiQiJianPi Formula (11.59 g/kg/d), the middle dose of YiQiJianPi Formula (23.18 g/kg/d) and the high dose of YiQiJianPi Formula (23.18 g/kg/d). In addition to the normal group of normal feeding, the other groups were eating feed containing 0.03% of ranitidine, and satiation two days and one day fasting. Except for model group given normal saline 2 mL by gavage, the other four groups each drug 2 mL, once a day. After 10 weeks, the general condition, weight change, viscera index change, pathology, RegI and PCNA were observed. Results: ①The mucosa of rats in model group was significantly thinner and glands decreased, with diffuse or focal lymphocytes, plasma cells and fibrous tissue, they showed moderate or severe inflammation. There were significant difference compared with the normal group. ②There were some changes in general condition and pathology treated by modern medicine and the different dose of YiQiJianPi Formula. The number of parietal cells in the low-dose group was more than the model group. The changes of the degree in gastric structural disorder were lighter than the model group. The gastric structures in the middle dose were more clearer and the epithelial cells arranged more regularer than the low-dose group. The gastric epithelial cells of high dose group were arranged neatly, similar to the control. ③There were no significant difference in organ index between different dose group and positive control group(P>0.05). However, it seemd to show the advantages of YiQiJianPi Formula from changes in value. ④The different dose of YiQiJianPi Formula and the modern medicine could decrease the contents of RegI and PCNA(P <0.05), while YiQiJianPi Formula were better than the modern medicine group(P< 0.05), and related to the dose. Conclusions:It can improve gastric pathology, organ index, the content of RegI and PCNA in serum and tissue of CAG rats, better the positive control group. We believe that the inhibition of the onset in CAG rats is related to the traditional Chinese medicine theory and composition of the drug-related external pharmacology, and correlated with the regulation of abnormal expression in Regl and PCNA. It may be one of mechanisms to treating CAG.This study reveals the part of the efficacy mechanisms for treating CAG. It will provide a scientific basis to confirme the efficacy for dominant diseases of TCM. |
