Objective:The aim of this study was to explore the role of insulin-like growth factor 1receptor(IGF-1R) and Livin in the formation and development of colorectal carcinoma and the relationship between IGF-1R and Livin by evaluating IGF-1R and Livin expression in colorectal carcinoma,colorectal adenoma and adjacent non-cancerous normal mucosa,looking forward to providing theoretic basis for that IGF-1R and Livin can be as effective biomarkers in evaluating the risk of carcinogenesis and the stage.Materials and Methods: A total of 60 primary colorectal carcinoma tissues(among 15Dukes’ A stage,15 Dukes’ B stage,15 Dukes’ C stage,15 Dukes’ D stage.) were collected from patients undergoing resection,30 colorectal adenoma tissues(among 8 tubular adenoma,22 villous tubular adenoma.) were obtained from patients undergoing colonoscopy examination and polypectomy,endoscopic mucosal resection(EMR) or endoscopic submucosal dissection(ESD),and 10 normal mucosa was approximately 5cm away from carcinoma tissues as control group.SP immunohistochemistry was utilized for expression analysis of IGF-1R and Livin in colorectal carcinoma,colorectal adenoma and adjacent non-cancerous normal mucosa.Results:1.Expressions of IGF-1R and Livin in colorectal carcinoma,colorectal adenoma and adjacent non-cancerous normal mucosa:The results showed the expression levels of IGF-1R in adjacent non-cancerous normal mucosa,colorectal adenoma and colorectal carcinoma gradually increased,and differences between the groups were statistically(all P value<0.05);the expression levels of Livin in colorectal adenoma andcolorectal carcinoma were signifiant high compared with adjacent non-cancerous normal mucosa(all P value < 0.05),however,the expression levels of Livin was no statistical significance between colorectal carcinoma and colorectal adenoma(P=0.889>0.05).2.The relationships between IGF-1R and Livin expression and clinicopathological characteristics in colorectal carcinoma:The expression level of IGF-1R had no association with sex,age,tumor location,tumor size,pathological types and tumor differentiation,but had strong association with invasion depth,Dukes’ stage,lymph node metastasis and distant metastasis(all P value < 0.05),and with the increase of invasion depth and Dukes’ stage and the occurrence of lymph node metastasis and distant metastasis,the expression level of IGF-1R was increased;the expression level of Livin had no association with sex,age,tumor location,tumor size,pathological types,tumor differentiation and invasion depth,but had strong association with Dukes’ stage,lymph node metastasis and distant metastasis(all P value< 0.05),and with the increase of Dukes’ stage and the occurrence of lymph node metastasis and distant metastasis,the expression level of Livin was increased.3.The relationships between IGF-1R and Livin expression and clinical characteristics in colorectal adenoma:The expression level of IGF-1R had strong association with the types of colorectal adenoma and cell atypia(all P value<0.05),the expression level in villous tubular adenoma was signifiant high compared with tubular adenoma,and with the increase of cell atypia,the expression level of IGF-1R was increased;the expression level of Livin had no association with the types of colorectal adenoma and cell atypia(all P value>0.05).4.The correlation of IGF-1R and Livin expression in colorectal carcinoma and colorectal adenoma:There was no correlation IGF-1R and Livin expression in colorectal carcinoma(rs=0.220,P=0.092) and colorectal adenoma(rs=0.000,P=0.999), and the differences were not statistically significant.Conclusions:1.There was a trend of increasing IGF-1R expression in colorectal adenoma and colorectal carcinoma,and high levels of IGF-1R expression suggest that IGF-1R played important roles in both the formation and development of colorectal carcinoma,it may be involved with the formation of malignant tumor in precancerous lesions by promoting the epithelial cells dysplasia.2.High levels of Livin expression has association with the formation of colorectal carcinoma,but may not be crucial facter in the transformation of colorectal adenoma to colorectal carcinoma.3. The expression level of IGF-1R had strong association with invasion depth,Dukes’ stage,lymph node metastasis and distant metastasis,and the expression level of Livin had strong association with Dukes’ stage,lymph node metastasis and distant metastasis suggested that IGF-1R and Livin may be as biomarkers in evaluating the condition of colorectal carcinoma.4.There was no correlation of IGF-1R and Livin expression in the formation and development of colorectal carcinoma.
