Objective:To observe the growth factor dynamic changes of acute myocardial ischemia rats and observe the effects of electroacupuncture on myocardial tissue.Methods:Sprague-Dawley Female rats were randomly divided into control (N), model (AMI). Use the left coronary artery ligation method make AMI model. Then AMI group were randomly divided into five group according to different observation time and a and electro-acupuncture group(EA). Observation indicators:1.Recording ECG; 2.Detected the expression of SCF, G-CSF and VEGF in blood serum by ELISA; 3.Detected the expression of SCF mRNA, G-CSFmRNA andVEGF mRNA in hippocampal by RT-PCR; 4.Detected the positive area CX43, TnI by Immune histochemical methodResults:1. The expression of SCF is on the rise, in 5 days present a peak, and there are differences in comparison with the normal group (P< 0.05);The expression of G-CSF in 5 days present a peak after acute myocardial infarction, in 4 days was a bit peak, compared with the N group have significant difference (P< 0.01), there are differences in 3days (P< 0.05); Present a tendency of increasing the expression of VEGF in the first day, and then fell down in 3 days after acute myocardial infarction (mi) with a low peak position, compared with the N group 1,4,5 days, the differences are statistically significant (P< 0.05).2.The expression of SCFmRNA-. G-CSFmRNA、VEGFmRNA present a downward trends after postoperation. SCFmRNA expression of the quantity of them in 5 days after acute myocardial infarction with a low peak,compared with the N group with statistical significance. (P< 0.05), CSFmRNA expression of 5 days after acute myocardial infarction toward a reduction in the peak, but compared with the N group there were significant differences (P< 0.01); VEGFmRNA expression of the rise in the first day, then fell down after 5 days with a low peak, than after 4 day compared with the N group with statistical significance. (P<0.05).3.Electric acupuncture group expression of CX43 positive was less than normal group and with statistical significance (P< 0.01), but the positive expression of electric acupuncture group area compared with myocardial infarction group have a significantly rise (P< 0.01);TnI positive expression area is increased in the normal group, and compared with normal group have a significant upward trend, and we have significant difference compared with normal group (P< 0.01).Electric acupuncture group and AMI group is a significant downward trend, and compared with AMI group had significant difference (P<0.01).4.The EA group of G-CSFmRNA, expression quantity were higher than in N group and AMI group, with significant differences (P< 0.01). The EA group of SCFmRNA, VEGFmRNA expression quantity were lower than in N group and AMI group with significant differences (P< 0.05); SCFmRNA, G-CSFmRNA Acute myocardial infarction group compared with normal group have SGF mRNA, SCFmRNA are down a bit, but no significant difference (P> 0.05), but for G-CSFmRNA expression decreased, compared with the N group there were significant differences (P<0.01).Conclusion:1. In acute myocardial infarction, cardiac muscle cell growth factor expression has certain change, has a rise in 1 day, and then gradually decline, VEGF decreased more significantly, and a sustained rise in serum of G-CSF, SCF have no difference compared with normal.2. Acupuncture can promote the expression of G-CSF, inhibition the expression of VEGF、SCF.3. Acupuncture can adjust the CX43, TnI have certain protective effect on acute myocardial infarction.
