Objective: To compare the expressions of cytokines HIF-1α,VEGF and PCNA in degenerative human intervertebral disc,and to explore the correlation of between disrupt annular fibrosus and intact annular fibrosus. Used to correlation of HIF-1α,VEGF and PCNA in disrupt annular fibrosus and intact annular fibrosus.Methods: The intervertebral discs originated from 42 cases receiving posterior approach excision due to the herniation of lumbar intervertebral discs. 21 of them appeared intact annular fibrosus,whereas other 21 cases appeared disrupt annular fibrosus. Disc degeneration was detected by HE staing. SP immunohistochemistry assay were used to detect the expressions and correlation of HIF-1α,VEGF and PCNA in disrupt annular fibrosus and intact annular fibrosus.Result: HE staining showed that nucleus pulposus cells of disrupt annular fibrosus were significantly reduced(P<0.01), The expression of HIF-1α,VEGF and PCNA in nucleus pulposus of intact annulus fibrosus group and disrupt annulus fibrosus group has been found. The expression of HIF-1α,VEGF and PCNA in disrupt annular group was significantly higher than those in intact annular fibroses group(P<0.01). HIF-1α expression was positively correlated with VEGF expression and PCNA expression was positively correlated with VEGF expression in both groups.Conclusion: The decrease of cell density is involved in the development of human Lumbar disc degeneration. The disc degeneration degree of disrupt annular fibroses group was severer than that of intact annular fibroses group. With the progression of disc degeneration, the expression of HIF-lα,VEGF and PCNA in the human degenerated intervertebral disc nucleus pulposus tissue gradually increase. Both HIF-1α and VEGF, VEGF and PCNA have a role in the course of intervertebral disc degeneration. It is presumed that HIF-1α and VEGF has an important role in the course of inhibiting intervertebral disc degeneration.
