Objectives By observing the changes of serum NO in rats with acute pancreatitis and the effect of Xuebijing Injection on it, it will investigate the role of NO in the AP and the possible mechanism of Xuebijing for AP treatment.Methods The 90 SD rats were randomly divided into 3 groups, they were sham operation group, acute pancreatitis group and Xuebijing group.Then each group was further divided into 3h, 6h and 12 h subgroups, 10 rats in each group. In the sham group,cut the abdomen of rats and did a simple flip pancreas; the AP group received laparotomy,through retrogradely biliopancreatic duct injection of 5% sodium taurocholate to set up the AP model; the Xuebijing group received Xuebijing injection treatment on the base of AP group. To observe the change of serum amylase(AMY), interleukin-6(IL-6), tumor necrosis factor alpha(TNF-α), nitric oxide(NO) and to observe the gross specimen pancreatic changes, the blood was taken at each time point and pancreas tissues were stained with HE for pathological examination.Results 1 Compared with the sham operation group, the pathological scores significantly increased at 3h, 6h and 12 h in the AP group, there was a significant difference between the sham group and the AP group(P<0.05). With the time prolonging,the pathological scores significantly increased in the AP group after 3h, 6h and 12 h, there was a significant difference among the AP group after 3h, 6h and 12h(P<0.05).Compared with the AP group, the pathological scores decreased obviously at 3h, 6h and12 h in the Xuebijing group, there was a significant difference between the AP group and the Xuebijing group(P<0.05). 2 Compared with the sham operation group, the content of serum AMY, TNF-α, IL-6 and NO significantly increased at 3h, 6h and 12 h in the AP group, there was a significant difference between the sham group and the AP group(P<0.05). With the time prolonging, the content of serum AMY, TNF-α, IL-6 and NO significantly increased in the AP group after 3h, 6h and 12 h, there was a significant difference among the AP group after 3h, 6h and 12h(P<0.05). Compared with the AP group, the content of serum AMY, TNF-α, IL-6 and NO decreased obviously at 3h, 6h and 12 h in the Xuebijing group, there was a significant difference between the AP group and the Xuebijing group(P<0.05).Conclusions 1 NO may be involved in the development course of acute pancreatitis and it is closely associated with the disease severity of acute pancreatitis. 2 Xuebijing injection can significantly reduce the content of serum NO and reduce the damage of pancreatic tissue.
