| Objective Dryopteris crassirhizoma Nakai, which is the dry root and petiole residue of Dryopterideceae of Dryopteris Adanson is widely used in China. According to Shen Nong’s Herbal, the efficacy of Dryopteris crassirhizoma Nakai is to treat the headache of wind-heat, the maculae of febrile virulent and sore swollen poison thanks to its cold in property, bitter in flavor and slight toxicity. Nowadays, on clinical, when it comes to curing cancer, including flu and fungal infection, it can be used in single or together with other herbs and it is recorded in China Pharmacopoeia. There are only a few of passages reveal that Dryopteris crassirhizoma Nakai is active in anti-fungi activity. But in our lab, we have already knowed thet phloroglucinol compounds are effective in anti-fungi activity. Nowadays, no report has showed that someone has done some studies on anti-fungi activity of Dryopteris crassirhizoma Nakai using its phloroglucinol compounds. So it is creative and valuable for us to do so, then test anti-fungi activity among its phloroglucinol compounds, summary our conclusion in the end.Methods In order to make a progress in studying on Dryopteris crassirhizoma Nakai, especially in exploring its constituents, repeated column chromatography on silica gel, Sephadex LH-20 and preparative should be applied. To learn how powerful is its phloroglucinol compounds on anti-fungi activity, Microdilution method from M38-A3 of Microsporum gypseum and Trichophyton rubrum should also be used in our study.Results 16 compounds were isolated in petroleum ether extraction of Dryopteris crassirhizoma Nakai through the way of our method. Their chemical structures were confirmed by their physical chemical constants, chromogenic reaction and spectroscopic analysis in the end. These compounds were(1)(8-hydroxymethylen)-trieicosanyl acetate,(2)AemulinBB,(3)Dryocrassin ABBA,(4)Filixic acid ABA,(5)Aspidinol B,(6) Cholesta-4-en-3-ol,(7)Chalinasterol,(8)Daphnoretin,(9)L-threosphinganine and(10) β-sitosterol,(11)Albaspidin AP,(12)Filixic acid ABB,(13)Phloropyron BB,(14) Friedelan-3,4-lactona,(15)Dryofragin,(16)1-Chlorotetradecane-2-ol.In our test, anti-fungi activity of compound 2, 3, 4, 5 were also studied. The anti-Microsporum gypseum activity of compound 2 is the best amog them, whoes MIC was 35μg/m L, others were 48.75μg/m L. We could conclude one rule from our study, that is dicyclic phloroglucinols are better in anti-fungi activity.Conclusion We could finally conclude that compound 1, 5, 6, 7, 8, 9, 13, 14, 15,16 where firstly isolated from this herb. Compound 2, 3, 4, 5 are effective in anti-fungi activity test. According to the research about the chemical constituents and anti-fungi activity of Dryopteris crassirhizoma Nakai, the further developments were laid by a solid theoretical foundation. |
