This dissertation included two parts: summary of related literatures ; studied on the chemical constituents, pharmacological activities, the content of dryocrassinABBA and HPLC fingerprint of effective parts from Dryopteris crassirhizoma. Part one: The documents about the present research situation in Chinese medicine Guan Zhong were summed up and analyzed. A review on the research progress of Dryopteris plants in chemical constituents and pharmacological activities was presented. 115 references were cited. Part two: According to the research on the commercial Guan-Zhong, we choose Dryopteris crassirhizoma as material to study, because it is the most popular plant used as Guan-zhong and it was recorded in Chinese pharmacopoeia. In this part, the chemical studies on Dryopteris crassirhizoma was closely linked to the pharmacological effectiveness. The qualitative and quantitative analysis of the anti-malarial fractions(AMF) were undertaken. 40 commercial Guan-Zhong samples collected from different provinces and districts were identified. It has become clear that Dryopteris crassirhizoma, D. peninsulae, Lunathyrium acrostichoides, Woodwardia japonica, W. unigemmata, Blechnum oriental and Malteuccia struthiopteris are used as Guan-Zhong at present.We found that Dryopteris crassirhizoma has anti-malarial activity for the first time. The whole methanol extract of Guan-Zhong and its 4 parts with different polarity were tested for screening their anti-malarial activity and anti-hepatitis B virus activity. The results showed that part A and part B of the extract possessed relatively strong anti-malarial activity and part A and part C can inhibit the secretion of HBsAg and HBeAg in cell line 2215 at 100μg/ml. The chemical constituents of Dryopteris crassirhizoma were investigated intensively for the first time. 28 compounds and an oil were isolated from the 4 parts of extract of Dryopteris crassirhizoma by silica gel, polyamide, sephadex LH-20 column chromatography. On basis of their spectral data, 20 of them were elucidated as hexacosanoic acid, filixic acid ABA, dryocrassin ABBA, nerolidol, pentacosanol, 3-hydroxy-heptacosanol, flavaspidic acid AB, trisflavaspidic acid ABB, albaspidin AA, hydroxyhopane, sucrose, β-D-fructopyranose, α-D-fructofuranose, β-D-fructofuranose, [2R,3R]-2,3-cis-3,5,7,3',5'-pentahydroxyflavan- -8-C-α-D-glucopyranoside ,caffeic acid and [2R,3R]-2,3-cis-3,5,7,3',5'--pentahydroxyflavan. Among them, trisflavaspidic acid ABB and [2R,3R]-2,3-cis-3,5,7,3',5'-pentahydroxyflavan-3-ol-8-C-α-D-glucopyranoside are new compounds and hexacosanoic acid, nerolidol, pentacosanol, 3-hydroxy-heptacosanol, flavaspidic acid AB, sucrose, β-D-fructopyranose, α-D-fructofuranose and β-D-fructofuranose, 1,3-di(3-hexinoic acid)-glyceride, 1-(acetylene-acetic acid)-3-(3-hexinoic acid)-glyceride, 1-(3-hexinoic acid)-glyceride, caffeic acid and [2R,3R]-2,3-cis-3,5,7,3',5'- -pentahydroxyflavan were obtained from Dryopteris crassirhizoma for the first time. The chemical constituents of the oil were analyzed by GC-MS and 12 components were identified as butyl-cyclohexane, cis-decahydro-naphthalene, 1-methylethyl-cyclohexane, 9-(1-methylidene)- -bicyclo[6.1.0]nonane, cyclohexyl-cyclohexane, (-)-3,7,7-trimethyl-11-methylene-spiro[5.5]undec-2-ene, [1S-(1α,3aβ,4α,8aβ)]-decahydro-4,8,8-trimethyl-9-methylene-1,4-methanoazulene, 3,7,11-trimethyl-2,6,10-dodecatrien-1-ol, caryophyllene, 2,3,4,4a,5,6-hexahydro-1,4a-dimethyl-7- (1-methylethyl)-naphthalene, [1aR-(1aα,7α,7aα,7bα)]-1a,2,3,5,6,7,7a,7b-octahydro-1,1,7,7a- tetramethyl-1H-cyclopropa[a]-naphthalene and α-curcumene. Another new compound, dryocrassin ABAA,was found by LC/MS/MS from AMF and dryocrassin ABPA was identified from dryopteris crassirhizoma for the first time. So 3 new compounds were found, 14 compounds were obtained from dryopteris crassirhizoma for the first time and 13 compounds were identified from dryopteris crassirhizoma for the first time.
