Objective To detected the expression of HIF-1α 、 VEGF and EGFR in esophageal squamous-cell carcinoma tissue, to explore the correlation of the three genes expression level and histilogical grade、invasive depth、regional lymph node metastasis, and explore the relationship of the three genes expression level and radiosensitivity in esophageal squamous-cell carcinoma tissue.Methods Protein expression of HIF-1α 、 VEGF and EGFR in 62 cases of esophageal squamous-cell carcinoma tissue and 10 cases normal esophageal tissue was detected by immunohistochemistry, to analysis the correlation of the three genes expression and histilogical grade、invasive depth、regional lymph node metastasis. 62 cases of initial treatment patients with esophageal squamous-cell carcinoma were treated by static intensity-modulated radiation therapy(IMRT), the dose of 95% was 59.4Gy. 62 case patients had received chest CT and esophageal imaging in 1-month after treatment, to determine the correlation of the three genes expression and recent radiotherapy effect.Results In esophageal carcinoma tissue, the positive expression of HIF-1α was 53.2%(33/62), mainly in cytoplasm or cell borders, the expression was significantly increased in tumor necrosis tissue surrounding. The positive expression of VEGF was 46.8%(29/62), mainly in cytoplasm; The positive expression of EGFR was 69.4%(43/62), mainly in menbrane. The positive expression of HIF-1α 、VEGF and EGFR were respectively 10%(1/10)、20%(2/10)、20%(2/10) in 10 cases normal esophageal tissue. The positive protein expression of the three genes in carcinoma tissues were significantly higher than that of normal tissue(p<0.01). The protein expression of HIF-1α 、VEGF and EGFR has a significant positive correlation with lymph node metastasis(N staging)(p<0.05), and has no obvious correlation with squamous cell carcinoma histilogical grade and invasive depth(p>0.05). In esophageal carcinoma tissue,the negative protein expression of HIF-1α was 29 cases, and 28 cases were radiation effectively, the effective rate was 96.6%(28/29); the positive protein expression of HIF-1α was 33 cases, and 21 cases were radiation effectively, the effective rate was 63.6%(21/33); there are statistically significant between the positive and negative expression of HIF-1α(X2 = 10.092;P = 0.002). In esophageal carcinoma tissue,the negative protein expression of VEGF was 38 cases, and 32 cases were radiation effectively, the effective rate was 84.2%(32/38); the positive protein expression of VEGF was 24 cases, and 19 cases were radiation effectively, the effective rate was 65.5%(19/24); there are no statistically significant between the positive and negative expression of VEGF(X2 = 0.256; P = 0.613). In esophageal carcinoma tissue,the negative protein expression of EGFR was 43 cases, and 33 cases were radiation effectively, the effective rate was 76.7%(33/43); the positive protein expression of VEGF was 19 cases, and 9 cases were radiation effectively, the effective rate was 47.4%(9/19); there are no statistically significant between the positive and negative expression of VEGF(X2=0.865,P=0.203).Conclusion There was a positive correlation between the positive protein expression of HIF-1α、VEGF、EGFR and lymph node metastasis in esophageal squamous-cell carcinoma tissue, the protein expression can stimulate the lymph node metastasis; The esophageal squamous-cell carcinoma carcinoma patients of positive overexpression of HIF-1α had a lower radiosensitivity, the expression level of HIF-1α can be used as a index to predict esophageal cancer recent radiotherapy effect.
