Burdock Fructooligosaccharide as a kind of functional fructooligosaccharide has attracted more and more attention. It is a excellent water soluble dietary fiber, low calorie, non digestion stomach acid and enzyme digestion, can directly reach the large intestine, selective absorption by Bifidobacterium, Lactobacillus, promote the proliferation of strains, produced short chain fatty acid and lower the intestinal pH, inhibit the growth of harmful bacteria, adjust the intestinal balance, and reduce the formation of harmful substances, protection of intestinal mucosal cell.This research through the growth state of strain and the production of short chain fatty acid isolated Lactobacillus plantarum can efficient metabolism of Burdock Fructooligosaccharide GF13.With the high fat diet combined with STZ to establish mouse models of type 2 diabetes, respectively intragastric administration of Lactobacillus plantarum, Burdock Fructooligosaccharide GF13 and Lactobacillus plantarum and Burdock Fructooligosaccharide GF13 combined preparations, for 4 weeks. Regular inspection of mouse body weight, fasting blood every week. After 4 weeks of treatment test fasting blood glucose tolerance with Roche glucose meter;ELISA kit for detection of serum lipids, insulin and inflammatory factors and other related biochemical indexes. Respectively, before treatment, treatment for 2 weeks, 4 weeks of treatment, stop treatment for 1 weeks used coated plate count method detected the number of Lactobacillus, Bifidobacterium, Enterobacteria, Enterococcus and Clostridium perfringens in mice intestinal contents; detected mainly short chain fatty acid content in intestinal contents after treatment.In the same way establish rat models of type 2 diabetes, testing the levels of gut barrier related factors such as D-Lac, NOS, ZON and DAO in serum.This study obtained the following conclusions:1. This study successfully screened 10 strains efficient metabolism of Burdock Fructooligosaccharide GF13 from 200 Lactobacillus plantarum strains which OD value reached 0.70~1.21 after 12 h culture medium, pH value of medium reached 5.37~5.77. Determination the yield of short chain fatty acids of the 10 strains L. plantarum in vitro fermentation, the yield of lactic acid during 1.76~2.29mg/mL, acetic acid productionv during 1.74~2.03mg/mL. The use of Sc52 on the Burdock Fructooligosaccharide GF13 was the highest, and the production of short chain fatty acids in the fermentation also was the highest, and laid a solid foundation forimproving the effect of Burdock Fructooligosaccharide GF13 combined with L. plantarumon type 2 diabetes.2. This study evaluated the L. plantarum Sc52 and Burdock Fructooligosaccharide GF13 used alone or combinated on the effect of the improvement and treatment of high fat diet in combinated with STZ induced mouse models of type 2 diabetes mellitus. L. plantarum Sc52 and Burdock Fructooligosaccharide GF13 alone or combined use can significantly reduce fasting blood of mice, improve the glucose tolerance, lower serum cholesterol, regulating lipid metabolism, reduce plasma endotoxin、proinflammatory cytokines in serum, thereby improving the low-grade inflammatory stateand insulin levels. L. plantarum Sc52 and Burdock Fructooligosaccharide GF13 combined treatment effect of the type 2 diabetes is better than alone.3. This study evaluated the L. plantarum Sc52 and Burdock Fructooligosaccharide GF13 used alone or in combination with improve effects of high fat diet in combination with STZ induced type 2 diabetes model mice intestine. Burdock Fructooligosaccharide is a kind of non digestible oligosaccharides, digested and utilizated by L. plantarum Sc52 in the intestinal tract, and can adjust the occupying colonization of related microbes in the gut.After treatment significantly increased the number of Lactobacillus, Bifidobacterium two main probiotics intestinal tract, form a biological barrier together with other anaerobic bacteria, inhibit the colonization of Enterobacterium, Enterococcus and other pathogenic bacteria, adjust the intestinal microbial balance. The added intestinal microflora fermentation to produce large amounts of short chain fatty acids in the colon, regulating blood lipid metabolism.L. plantarum Sc52 and Burdock Fructooligosaccharide GF13 combined treatment effect of the type 2 diabetes is better than alone.4. This study evaluated the L. plantarum Sc52 and Burdock Fructooligosaccharide GF13 used alone or in combination with improve effects of high fat diet in combination with STZ induced type 2 diabetes model rats intestinal barrier function improvement. Mainly by improving the intestinal barrier function in damaged state, repair damaged cell membranes, reduce levels of intestinal barrier cytokines DAO and D-lac, NOS, ZON in rats serum, reduce the intestinal permeability, raise the level of GLP-2 and ADPN, maintaining the integrity of cell tight junction, improve immune function, reduce the level of serum inflammation factor TNF-a, IL-1, IL-6, IL-1β; promote the secretion of mucin, reduce and inhibit the adhesion of the pathogen and endotoxin, thereby improving the low-grade inflammatory state and insulin levels.L. plantarum Sc52 and Burdock Fructooligosaccharide GF13 combined treatment effect of the type 2 diabetes is better than alone.
