ObjectiveMissed abortion is a special type of abortion, it could bring great pain to the patients and their family. The exact pathogenesis of abortion is not fully understood. In this study, the expression of thrombospondin-1(TSP-1) and its receptor CD36 in serum and decidua of normal early pregnant women and patients with missed abortion were measured, and the expression of CD34 in decidua was measured, also the microvessel density(MVD) in decidua were studied, simultaneously the expression differences of them were compared. By exploring relationship between abnormity of TSP-1 and CD36 expression and missed abortion, the correlation between TSP-1, CD36 and MVD were discovered. The thesis could provide a theoretical basis for the study of causes and pathogenesis of missed abortion. Method30 cases of missed abortion as the case group and 30 cases of normal early pregnant women as the control group who were required to induce abortion in gynaecology and obstetrics of Affiliated Hospital of Yan’an University were chose to study in May 2014 to October. Before the artificial abortion, the serum of 60 cases of above-mentioned patients was collected and OD value of TSP-1 and CD36 in serum were detected by enzymelinked immunosorbent assay(ELISA), and their serum concentration were calculated. After the artificial abortion, decidua tissues of 60 cases of patients were collected for pathological examination, meanwhile, streptavidin-biotin peroxdase(SP) was employed to detect the expression of TSP-1, CD36 and CD34 in decidua. The results of immunohistochemistry were semi-quantitatively calculated by H-score method and decidual MVD value was counted. The data were input into SPSS19.0 software for statistical analysis. ResultsThe serum of the case group and the control group were both detected TSP-1 and CD36, The content of TSP-1 and CD36 in the serum of the case group were more than the control group. The differences were statistically significant(P<0.05); The expression of TSP-1 and CD36 in serum in the case group and the control group showed a positive correlation, The differences were showed to be significant(P<0.01); TSP-1 and CD36 in decidua of the two groups are positive expression and the areas of positive expression are mainly distributed in the cell membrane and cytoplasm of decidua, the H-score of TSP-1 and CD36 in decidua in the case group were higher than the control group, there are significant differences between the above two groups(P<0.05); The expression of TSP-1 and CD36 in decidua in the two groups showed a positive correlation. Their differences were showed to be significant(P<0.05); The MVD value in decidua of the case group were lower than the control group, the differences were significant(P<0.01); TSP-1 and CD36 in decidua and serum of the case group are positive expression(P<0.05). In decidua and serum, the expression of TSP-1 and CD36 in the control group were both not significantly related(P>0.05); In decidua, the expression of TSP-1 and MVD in the two groups showed negative correlation(P<0.05). ConclusionThe high expression of CD36 and TSP-1 in the serum and decidua and the decrease of MVD in decidua were related to incidence of missed abortion. TSP-1 and MVD in decidua were negatively correlated. The high expression of TSP-1 and CD36 in the local microenvironment of decidua may play the role of inhibition of angiogenesis, angiogenesis were inhibited and the balance of the vascular system in decidua were destroyed by the combination of TSP-1 and CD36, The change could reduce angiogenesis in decidua and impact its function, interfere implantation of embryos and the maintenance of pregnancy, lead to make embryo insufficiency and hinder the development of embryos until the missed abortion occur. In decidua and serum, the expression of TSP-1 and CD36 in normal early pregnancy group were both not significantly related, TSP-1 and CD36 in decidua and serum of patients with missed abortion were positive expression, so we guess the high expression of TSP-1 and CD36 in decidua of patients with missed abortion could express in peripheral blood, so that monitoring the content of TSP-1 and CD36 in the serum of the early pregnant women can be used as an auxiliary diagnosis of missed abortion, but not for the exact diagnosis of the disease, which is of limited significance to the early detection of missed abortion.
