Objective Detecting the Expression strength of osteopontin and vascular endothelial growth factor in normal endometrium,normal myometrium, eutopic and ectopic endometrium with adenomyosis.In order to explore the the role of OPN and VEGF and their relationship in the development of adenomyosis.Methods1, Selected 40 patients with adenomyosis as the study group.Cut the eutopic and ectopic endometrium tissues as the speciman as long as the patients finished an panphysterectomy.2, Selected 20 patients with uterine myoma as controls,Cut the normal endometrium and myometrium tissues as the speciman as long as the patients finished an panphysterectomy.3, Using the Two-steps Method of Immunohistochemistry to detect the OPN and VEGF in innormal endometrium,normal myometrium,eutopic and ectopic endometrium with Adenomyosis.Then using the Leica Q500 MC image analysis system to dectect the intensity of OPN and VEGF expression.4, Using SPSS13.0 statistical software to analyze the results.The positive expression was described in a form of positive unite(PU)with mean±standard deviation.Results1, The positive expression of OPN and VEGF were observed in the cytomembrane of normal、eutopic and ectopic endometrial gland cells,and also in the myometirum cells. OPN and VEGF were expressed in both adenomyosis lessions and normal myometirum.Their PU of OPN in the two group were 27.15±9.05 and 11.64±4.49, while the PU of VEGF were36.92±6.14 and 15.63±3.93.Accroding to the T-test,the difference was statistical significance(P <0.05).2,To adenomyosis,the expression of OPN in secretory phase was higher than proliferative phase.Their PU were 25.12±6.85 and 19.89±8.15,the difference was statistical significancel( P <0.05).To myometruim,the expression of OPN in secretory phase was higher than proliferative phase.Their PU were12.41±4.96 and 13.12±3.48, the difference was not statistical significance(P >0.05).The PU of OPN in proliferative phase 、 secretory phase of ceutopic group were30.19±4.79 and 28.59±4.42. Comparing to the controls 19.89±8.15 and 13.12±3.48,there made significance P <0.05.3, To adenomyosis the expression of VEGF in secretory phase was higher than proliferative phase.Their PU were 30.19±4.79 and 28.59±4.42,the difference made no sense(P >0.05).To myometruim,the expression of VEGF in secretory phase was higher than proliferative phase.Their PU were 17.28 ± 6.64 and 14.55±3.21 the difference was not statistical significance(P >0.05).The PU of VEGF in proliferative phase 、 secretory phase of ceutopic group were30.19±4.79 and 28.59±4.42. Comparing to the controls 17.28±6.64 and 14.55±3.21,there made significance P <0.05.4, According to Pearson correlation analysis,in adenomyosis lesions,OPN and VEGF had not correlation with each other.Conclusion1, The expression of OPN and VEGF in eutopic and ectopic endometrium with Adenomyosis were higher than normal endometrium. It idicated that these two factors were involved in the development of adenomyosis.2,In eutopic group, OPN and VEGF all had high degree expression in proliferative phase、secretory phase,prompt that cells in the two stage endometrial have a more powerful ability to invade the myometrium and lead to the adenomyosis.Meanwhile the expression of OPN and VEGF in secretory phase was higher than proliferative phase,showed that comparing to proliferative phase,cells in secretory phase were more easy to invade the myometrium.3,OPN and VEGF had not correlation with each other in adenomyosis.Which indicated thatboth factors have indepenent regulations in the progress of adenomyosis.
