Objective: To investigate the mechanisms and effects of “Cuyushengjingfang” on Cat Sper1 and Cat Sper2 in the sperm of Oligoasthenozoospermia Wistar rats induced by Cyclophosphamide(CP), to provide scientific basis for the clinical applications.Methods: Fifty male Wistar rats were acclimated for six days, then divided randomly into model group(MG), blank control group(BCG), low-dose Cuyushengjing group(LDCG), middle-dose Cuyushengjing group(MDCG) and high-dose Cuyushengjing group(HDCG), 10 rats in each group. Model rats of oligoasthenozoospermia were induced by CP(30mg/kg) injected into the abdominal cavity of rats once a day for 5 days, while BCG were injected with normal saline(NS). The rats in LDCG, MDCG and HDCG were oral administrated with 3ml Cuyushengjingfang for 30 days while the rats in MG and BCG were oral administrated with 3ml NS. The concentration of Cuyushengjingfang in that of LDCG, MDCG and HDCG were 1.73mg/ml, 3.47mg/ml, 6.93mg/ml. Then sperm amount were measured by light microscope. The Ca2+ inner the sperm were detected by floe cytometry. The m RNA and protein expression of Cat Sper1 and Cat Sper2 in sperm were measured by PCR and Western blot.Results: Models rats of Oligoasthenozoospermia were created successfully. By observing the number of sperm and the sum of a+b degree sperm rate, the m RNA and proteins expression of Cat Sper1 and Cat Sper2, the Ca2+ inner the sperm, compared with BCG group, MG showed obvious decrease(P<0.05).Compared with MG, the number of sperm and the sum of a+b degree sperm rate, the m RNA and proteins expression of Cat Sper1 and Cat Sper2, the Ca2+ inner the sperm were increased significantly in MDCG and HDCG(P<0.05).Conclusion: Cuyushengjingfang could effectively increase the sperm amount and the expression of Cat Sper1 and Cat Sper2 in the model Rats of Oligoasthenozoospermia created by Cyclophosphamide, so that the Ca2+ inner the sperm were increased and sperm quality were increased. |
