Objective:To build oligo/asthenospermia rat model and to discuss changes of sperm quality, testicular pathological morphology, and cell apoptosis and energy metabolic pathways’ protein and gene expression.Methodology:Divided 70 experimental rats, weight 300-330g, into 7 groups:normal group, model group, Huangjing Zanyu high-medium-low dose groups, Wuziyanzong pill control group and 1-carnitine control group. Build oligo/asthenospermia model induced by cyclophosphamide and appropriate drug intervention. After the test, using semen analysis to examine sperm quality; light microscopy to observe testicular pathological morphology; and western blot and Real-time fluorescent quantitative PCR to detect and analyze the protein and gene expression of cell apoptosis pathway Pi3k-Akt and energy metabolic pathway Catsperl.Results:1. Demonstrated that cyclophosphamide was used successfully in the development of the oligo/asthenospermia rat model by observing the general condition, sperm routine and testicular pathological morphology observation of the rats.2. Semen examination results showed that after the intraperitoneal injection by cyclophosphamide, the experimental rat’s B grade sperm percentage, forward sperm (PR), and sperm activity rate were significantly reduced. Following Huangjing Zanyu capsule intervention, these three indicators displayed significant improvement. The quality of oligo/asthenospermia rats’sperm was improved.3. Testicular tissue biopsy showed pathological changes following intraperitoneal injection of cyclophosphamide; the experimental rats’testicular tissue and spermatogenesis were compromised. Huangjing Zanyu capsule repaired damaged testicular tissue and enhanced oligo/asthenospermia rats’ spermatogenesis resulting in improved sperm quality.4. The intraperitoneal injection of cyclophosphamide reduced the protein and gene expression of cell apoptosis Pi3k-Akt pathway (Pi3k segment) and energy metabolic pathway Catsperl. After Huangjing Zanyu capsule intervention, both were improved, thereby increasing the number and motility of sperm.Conclusion:Huangjing Zanyu capsule can significantly improve the quality and quantity of sperm in oligo/asthenospermia rats and repair damaged testicular tissue. It may be related to increasing the protein and gene expression of cell apoptosis Pi3k-Akt pathway (Pi3k segment) and energy metabolic pathway Catsperl. This can provide molecular level experimental evidence of Huangjing Zanyu capsule’s mechanism in the improvement of weak sperm.
