Objective:To investigate the effect of post-conditioning with Propofol on apoptosis and the expression of bcl-2,bax gene in renal tissue after hepatic ischemia-reperfusion injury and its mechanism. Methods:Fifty-four healthy male SD rats were randomly divided into three groups, including sham group(S), ischemia-reperfusion group(IR), propofol group(P).According to the different periods after reperfusion, each group was further divided into 2-hour 4-hour and 6-hour reperfusion subgroups respectively(n=6 in each subgroup), named S2、S4、S6 subgroups,and IR2、IR4、IR6 subgroups, and P2、P4、P6subgroups. Make the 70% hepatic ischemia-reperfusion model. Except the S group, other groups experienced liver ischemia of 45 minutes and then recovered reperfusion. S group and IR group was pumped 0.9% saline(0.3ml·kg-1·min-1) 10 minutes after liver ischemia of 35 minutes. P group was pumped 0.1%propofol(0.3ml·kg-1·min-1) 10 minutes. The left kidney were sampled at the end of reperfusion and observed by morphology and hematoxylin and eosin(HE) staining.The blood was sampled at the end of ischemia-reperfusion for assay of cysteine proteinase inhibitor(Cys-c)the apoptosis rate(AI) in ischemia-reperfusion renal tissure were measured by TUNEL method; the Bcl-2,Bax concentration were measured by immuno-histochemistry. Results:1.Compared with S group, the expression of Bcl-2 and Bax protein increased both in group IR and E(P<0.05),with statistical significance. Compared with IR group, the expression of Bax protein decreased and Bcl-2 protein increased in group P(P<0.05),with statistical significance.2.Tunel staining showed:compared with S group, the AI increased both in group IR and P(P<0.05, with statistical significance. Compared with IR group, the AI decreased in group P(P<0.05),with statistical significance.3.Compared with S group, the content of Cys-c increased both in group IR and P(P<0.05),with statistical significance. Compared with IR group, the content of Cys-c decreased in group P(P<0.05),with statistical significance.4.The pathological lesions were lighter in group P than that in group IR under light microscope. Conclusion:1. Propofol can alleviate renal injury induced by hepatic ischemia-reperfusion in rats.2. The bcl-2 and bax protein may play important roles.
