Objective:The aim of this study was to observe the change of circulating betatrophin in diabetic patients by examine circulating betatrophin concentrations in patients with type 1 and type 2 diabetes, and to analysis the relativity between serum betatrophin levels and clinical of indicators. Methods:A total of 154 subjects(80 healthy controls, 30 type 1 diabetes,44 type 2 diabetes) were enrolled in the study. The indexes of body and blood biochemical indexes of metabolism were tested, and Circulating betatrophin was measured with ELISA kit,and the relativity between betatrophin levels and above indexes were analyzed by statistical method. Results:Fasting blood glucose, blood glucose 2 hours after meal, glycosylated hemoglobin were significantly increased in patients with type 1 compared with healthy subjects. Fasting C peptide, C peptide 2 hours after the meal, HOMA-β were significantly reduceed in patients with type 1. In patients with type 1 diabetes, serum betatrophin concentration was increased compared with healthy subjects(3.57±0.28 ng/ml vs 2.18±0.24 ng/ml,p<0.01),and correlated with age, Hb A1 c, fasting blood glucose, fasting insulin, fasting C peptide and HOMA-β. HOMA-β, fasting blood glucose, age and Hb A1 c were the main influencing factors of Betatrophin by multiple linear regression analysis in patients with type 1 diabetes. Body mass, BMI, waist circumference, waist-hip ratio, triglycerides, fasting blood glucose, blood glucose 2 hours after meal, fasting insulin, insulin2 hours after meal, fasting C peptide, C peptide 2 hours after the meal, Hb A1 c, HOMA- IR were high in patients with type 2 compared with healthy subjects, and HOMA-β was contrary. In patients with type 2 diabetes, serum betatrophin concentration were significantlyincreased compared with healthy subjects(4.87±0.28 ng/ml vs 2.26±0.24 ng/ml,p<0.01),and there was a postive correlation between Betatrophin and age, BMI, WHR, fasting blood glucose, fasting insulin, Hb A1 c, cholesterin, triglycerides, low densith lipoprotein(LDL), HOMA-IR, and the Betatrophin was negatively related to high density lipoprotein(HDL), HOMA-β. BMI, HOMA-IR, HOMA-β, fasting insulin and age were the main influencing factors of Betatrophin by multiple linear regression analysis in patients with type 2 diabetes. In patients with type 2 diabetes, serum betatrophin concentration was higher than that of patients with type 1 diabetes(4.87±0.28 ng/ml vs3.57±0.28ng/ml,p<0.01).Conclusion:The serum betatrophin levels were elevated in patients with type 1 diabetes and type 2diabetes. serum betatrophin concentration was higher than that of patients with type 1diabetes.In patients with type 1 and type 2 diabetes mellitus, betatrophin was associated with HOMA-β, showing that betatrophin was associated with the function of islet beta cells. In patients with type 2 diabetes, betatrophin was associated with serum lipid parameter; While betatrophin was only associated with glucose metabolism in patients with type 1 diabetes. Betatrophin is involved in lipid metabolism. In conclusion, this study will provide theoretical basis for betatrophin in the treatment of diabetes.
