Citrullus colocynthis was also known as bitter gourd, bitter apple, bitter gourd, which is Cucurbitaceae. Citrullus colocynthis usually refered to its dried ripe fruit. Cucurbitaceae.Citrullus colocynthis is one of the traditional Uygur medicine, which was used as laxatives and diuretics in clinical practice and treat other symptoms of rheumatoid arthritis. Afterwords Citrullus colocynthis was also used for the treatment of liver diseases, blood diseases, diabetes and cancer. Citrullus colocynthis mainly grows in the desert of North African and southern Europe Central Asia area. Citrullus colocynthis contains a lot of bioactive constituents,such as triterpenoid saponins, flavonoids and their saponins, alkaloids, fatty alkanes, alcohols and other medicinal ingredients. In order to further study effective medicinal chemical composition of Citrullus colocynthis and make specific bioactive components more clear in this plant to better apply traditional drug to pharmacological experiments by various chemical components of modern clinical medical field, In this paper, the experimental research was followed as the following aspects.1. Research of chemical constituents from Citrullus colocynthis ethanol extractionnbutanol extraction(1) The powed Citrullus colocynthis was extracted by ethanol extracts and then the ethyl alcohol extraction was extracted by petroleum ether, chloroform, ethyl acetate, n-butanol.ethyl acetate and n-butanol fraction was sub Jected to silica gel column chromatography, MCI gel column chromatography, column chromatography, ODS Rp-18 column chromatography,preparative thin-layer chromatography plates to separate and purify the compounds. combined with modern spectral analysis techniques and physical and chemical properties, 13 compounds were identified: p-hydroxy benzoic acid, benzoic acid, phthalic acid ester,catechol, isophthalic diphenol, 4-hydroxybenzoate, 4-methylenecyclohexa-2,5-dienone, 2-O-β-D-glucopyranoside cucurbitacin I, 16-prop-1-en-yl-2-O-β-D-glucopyranosylcucurbitacin I,gastrodin, quercetin, 16-prop-1-en-2-yl-25-O-acetyl-2-O-β-D-glucopyranosylcucurbitacinI, 16α, 23-epoxy-2, 20β-dihydroxycucurbita-1, 5, 24-diene-3, 11-dione 2-O[α-1rhamnopyranosyl-(1â†'2)-α-D-glucopyranoside].(2) The new compounds of 16-prop-1-en- yl-2-O-β-D-glucopyranosylcucurbitacin I, 16-prop-1-en-2-yl-25-O-acetyl-2-O-β-D-glucopyranosylcucurbitacin I were reported firstly.2. Uygur medicine Citrullus colocynthis total flavonoids extraction and purification processed by macroporous resins researchuygur medicine Citrullus colocynthis total flavonoids extraction and purification processed by macroporous resins research: via single factor experiments and orthogonal experiments,with temperature, solid-liquid ratio, extraction time, solvent volume fraction as process parameters, and Citrullus colocynthis total flavonoids were extracted to determine the optimum process parameters under three extraction methods including reflux, microwaveassisted, ultrasound-assisted extraction method under different extraction conditions.(1) Reflux extraction optimum conditions of total flavonoids were that the temperature is90 oC, extraction time was 2h, the concentration of ethanol was 95%, solid-liquid ratio was1:80.(2) Microwave-assisted extraction of total flavonoids optimum conditions: the temperature was 90 oC, ethanol volume fraction of 95%, the extraction time for the extraction time was25 min, solid-liquid ratio was 1: 90.(3) The optimum conditions using ultrasonic assisted extraction of total flavonoids from Citrullus colocynthis: extraction temperature was 70 oC, ethanol concentration is 90%, the extraction time was 10 min, solid-liquid ratio was 1: 125.(4) The flavonoids extraction from Citrullus colocynthis was purified by macroporous resins. By comparing the eight kinds of macroporous resins’ adsorption and resolve performance to flavonoids of Citrullus colocynthis, D101 resin was the most suitable resin to purify the flavonoids compounds from Citrullus colocynthis. The optimum adsorption condition were that the volume flow was 3 m L/min, p H value was 6, with those conditions the D101 macroporous resin had the largest absorbing capacity that is 13.638 mg/g. Flavonoids optimum elute condition were that 5BV deionized water was used to eliminate impurity and the flavonoids was eluted with 95% methanol in 1m L/min flow rate.3. By comparing the eight kinds of macroporous resins’ adsorption and resolve performance to flavonoids of Citrullus colocynthis, D101 resin was the most suitable resin to purify the flavonoids compounds from Citrullus colocynthis. The results showed that: control volume flow to 3 ml/min, PH value is 6, the largest adsorption capacity triterpenoids D101 macroporous resin was 9.333 mg/g(dry resin mass calculation). Triterpenoids of optimum elute condition were that 5 BV deionized water was used to eliminate impurity and the flavonoids was eluted with 95% methanol in 1 ml/min flow rate.
