Shengmai Yin,made by the percolation with ethanol in Pharmacopoeia, has some problems as follows: long production cycle, ethanol residue, inconsistent quality and the backwardness of the quality evaluation methods,etc.. Therefore, the experiment was conducted with the HPLC method to detect the 16 kinds of ginsenoside monomers as the index and optimized the processing technology of Mai Wei Shen. The method of simultaneous determination of twenty active constituents in the Mai Wei Shen was established by HPLC. The Mai Wei Shen was evaluated on hypoxia tolerance, anti-fatigue, enhancing the immunity and serum lipids by the experiments in vivo with mice. The expreiments provide us with a “safety, effective, controllable ” theoretical basis and reference of Mai Wei Shen. Experimental results are as follows.1. The best processing tehnology for Mai Wei Shen was the ratio for ginseng, radix ophiopogonis and schisandra chinensis was one to three to one, used to radix ophiopogonis and schisandra chinensis with solid-liquid ration of one to ten, soaked for 30 min, boiled for 90 min, and decocted two times. The filtrate was merged and concentrated. Then brought the ginseng into the filtrate, and fried the filtrate with water bath by 100℃, dried it by 100℃, and crushed it through 200 mesh sieve.2. The method of simultaneous determination of 16 kinds of ginsenoside monomers, 3 kinds of schisandra lignin, and 1 kind of ophiopogon japonicus saponin in the Mai Wei Shen by high-performance liquid chromatography was developed.3. Concentration of ginsenosides F2, Protopanaxatriol, Compound K, Rh2 in Mai Wei Shen were higher than those in ginseng(P<0.05).The value added of 16 kinds of ginsenoside increaded nearly three times than ginseng, concentration of ginsenoside Rg1, Re, Rc, Rb3, Rd, Protopanaxadiol decreased, but higher than the decoction of ginseng(P<0.05),ginsenoside Rg3, which was not detected in ginseng, was detected in Wai Wei Shen with the content of up to 0.15%.4. compared with ginseng and Shengmai Decoction, Mai Wei Shen could significantly improve the survival time in hypoxia tolerance, sodium nitrite poisoning and acute cerebral hypoxia of mice(P<0.05), reduce the content of cyanide high-speed rail hemoglobin for sodium nitrite poisoning of mice(P<0.05), significantly increased the activity of SOD and GPX(P<0.05), and reduce the content of MDA in liver and brain tissue of mice(P<0.05). It has the function of auxiliary improving hypoxia tolerance.5. Mai Wei Shen could prolong the swimming time of mice(P<0.05), reduce the content of serum LBA, BUN of fatigue mice(P<0.05), and it could increased the content of the liver glycogen and muscle glycogen of mice(P<0.05), which has the effect of relieving sports fatigue.6. Mai Wei Shen could significantly increased the index of immune organs of mice(P<0.05), the ratio of T lymphocyte subsets of mice(P<0.05), IL-2 and TNF-α cytokine levels(P<0.05), the content of immunoglobulin of mice(P<0.05. Moreove, it has the significant effect of immunosuppression.7. Mai Wei Shen did not affect the content of total cholesterol, triglycerides, high density lipoprotein cholesterol and low density lipoprotein in blood of healthy mice(P>0.05).
