Objectives: To study the expression of p27 and Ki-67 in hepatocellular carcinoma (HCC) and the relationship among p27 and Ki-67 and clinic pathologic parameters, and to determinate whether the change of p27 occures by post-transcription mechanisms.Methods: The expression of p27 and Ki-67 were measured with immunohistochemistry assay (SP-method) in thirty-three HCC and ten normal liver tissues. Meantime, the reverse transcriptase PCR (RT-PCR) was carried out to detect the level of p27mRNA from the above mentioned 33 HCC and 10 normal samplesResults: p27 average optical density (AOD) was correlated with malignancy degree and progression of HCC. The AOD in HCC with good differentiation was obviously higher than that with poor differentiation(0.24?.06 vs 0.16 + 0.03, p<0.001). The AOD in HCC without carcinoma embolism was higher than that with carcinoma embolism(0.22 + 0.06 vs 0.14 + 0.02, pO.OOl) , and in HCC with diameter <3 cm was higher than those with diameter >3 cm(0.22+0.05 vs 0.16 ?.05, p<0.005), and in HCC in ? -?a stage was higher than that in HCC in ?b-?stage(0.22 + 0.05 vs 0.16 ? 0.05, p<0.01). No correlation was found between p27 AOD and patient's sex and serum AFP level (p>0.05). Furtherly, the Ki-67 index was correlated with malignancy degree and progression of HCC. The Ki-67LI in HCC with good differentiation was obviously lower than that with poor differentiation (176.5 + 32.3 vs 240.4 ? 97.7, p<0.05), the Ki-67LI in HCC without carcinoma embolism was lower than that with carcinoma embolism(164.6 ? 38.0 vs 283.9 ? 81.00, p<0.001) , the Ki-67LI inHCC with diameter <3 cm was lower than that with diameter >3 cm (172.1 ? 57.7 vs 239.9 ? 87.9, p<0.05), and in HCC of ? -?a stage was lower than that in HCC of ?b-?stage (162.1?38.3 vs 279.0? 79.8, p<0.001),and in HCC with serum AFP<20?g/L was lower than that in HCC with serum AFP?20?g/L (165.7?44.5 vs 233.8?88.4, p<0.05). The p27 AOD with Ki-67LI?200 was lower than that with Ki-67 LI <200 (0.16?0.04 vs 0.21?0.06, p<0.05). p27 mRNA levels was not significantly different from the levels observed in the corresponding samples.Conclusion: The expression of p27 and Ki-67 is corrected with invasiveness and progression of HCC. Changes of p27 in hepatocellular carcinoma may occur due to the post-transcription mechanisms.
