OBJECTIVE:Aim to study the effect of GCF2selected cutting in livercancer occurrence and development by detecting the expression of five selectedcutting zones of GCF2in normal liver cell lines, liver tissue, liver cancer lines,liver cancer tissues and Ovarian cancer cell lines.METHOD:according to Genbank database person GCF2six differenttranscripts, predicting GCF2genes five alternative splicing area (D1~D5);against sequence features D1-D5is designed and synthesized five pairs ofprimers used to detect RT-PCR D1~D5respectively, in normal liver cell lines,liver cancer cell lines and human ovarian cancer cell lines as well as43cases ofhepatocellular carcinoma/adjacent tissues, the expression of nine cases ofnormal liver tissue, and DNA sequencing of PCR products were positive.Andaccording to the sequencing results by bioinformatics predicted secondarystructure of the mRNA alternative splicing of GCF2.RESULT:(1) GCF2choice of four shear zones (D1~D4) are not expressed in the detected nine kinds of cell lines, alternative splicing area D5only positive expression in tumor cell lines;(2) D1~D4are not expressed inthe liver, cancer and adjacent normal liver tissues, D5tissues next to the41cases of liver cancer/cancer, are not expressed in the two cases of liver cancer/cancer tissues and nine cases of normal liver tissue adjacent.(3) In the tissueof skin、 ileum、 colon、tonsil、cervix、appendix、uterus、thyroid、kidney、muscle、rectus abdominis、ovary、fat、4cases of normal brain tissue and5casesof glioma tissue,D1~D44alternatively spliced region not expressed;alternatively spliced region D5expressed in tissue of skin, ileum, colon,appendix, kidney, there are three cases expressed in four cases of normal braintissue in5cases,4cases of glioma tissue expression; while in the tonsils,cervix, uterus, thyroid, muscle, rectus abdominis, ovary, adipose tissue did notexpress.(4) alternatively spliced region D1~D5mRNA free energy of thesecondary structure of the original were-22.0,-45.0,-31.7,-6.4,-138.2.CONCLUSION: GCF2choice of four shear zones (D1~D4) and tumordevelopment may no significant correlation; selective shear zone D5detectedin tumor cell lines, liver/paracancerous positive expression organizations, D1~D5original five splicing area D5free energy area, the most stable secondarystructure; splicing region D1~D4mRNA secondary structure are unstable,wherein the splicing region D4most unstable. Tips GCF2choice shear zone D5may occur, such as the development of liver cancer tumors.
